Sortilin restricts secretion of apolipoprotein B-100 by hepatocytes under stressed but not basal conditions

Donna M. Conlon et al.

Summary: Genetic variants at the SORT1 locus in humans are associated with reduced plasma levels of LDL cholesterol and apoB. Loss of Sort1 expression in mice leads to increased apoB secretion under lipid loading or ER stress conditions. Carriers of SORT1 variant with diabetes show larger decreases in plasma apoB, TG, and VLDL and LDL particle number. Hepatic sortilin may serve as a quality control step in the apoB secretion pathway.

Inceptor counteracts insulin signalling in β-cells to control glycaemia

Ansarullah et al.

Summary: The discovery of inceptor, an inhibitor of insulin receptor (INSR) and IGF1 receptor (IGF1R) signalling in mouse beta-cells, sheds light on a potential molecular target for INSR-IGF1R sensitization and diabetes therapy. The study shows that inceptor plays a role in shielding insulin-producing beta-cells from constitutive pathway activation, which could have implications for improving glucose tolerance in vivo.

Microtubules in insulin action: what's on the tube?

Skylar R. Batty et al.

Summary: Microtubules play a crucial role in insulin-stimulated glucose transport in adipocytes by facilitating the trafficking of GLUT4 vesicles through the cell. The integration of cytoskeletal networks and involvement of molecular motor proteins are essential for the complex movement of GLUT4 before its insertion into the plasma membrane for glucose transport. Insulin response and the regulation of MT-associated proteins are important factors in understanding the mechanisms of insulin action in adipocytes.

Summary: This study focused on the characteristics and optimization of GLP-1 receptor endocytosis, using various methods to analyze the effects and screen potential agonists for treatment of type 2 diabetes mellitus. The results showed that endocytosis of GLP-1 receptor-mediated elimination was clathrin-dependent, and agonists biased towards the G protein pathway had less endocytosis. The identified analogue M4 showed prolonged hypoglycaemic activities and a long half-life, making it a potential lead compound for type 2 diabetes mellitus treatment.

Revising Endosomal Trafficking under Insulin Receptor Activation

Maria J. Iraburu et al.

Summary: The endocytosis of ligand-bound receptors and their recycling to the plasma membrane are important for cell functions. The insulin receptor can be endocytosed by clathrin-dependent and independent mechanisms, and the sorting of the receptor at the early endosome determines the intensity and duration of insulin effects. Various proteins, including the Arf and Rab families of small GTPases, regulate the endocytic and endosomal pathways. This article argues for a specific role of the slow recycling route in INSR endocytosis and provides a review of the molecular mechanisms involved.
