Sensory Neurotransmitter Calcitonin Gene-Related Peptide Modulates Tumor Growth and Lymphocyte Infiltration in Oral Squamous Cell Carcinoma

Lisa A. McIlvried et al.

Summary: Head and neck squamous cell carcinoma are highly innervated by peripheral sensory neurons. Local neurotransmitter release, such as CGRP, from sensory neurons innervating cancer is linked to tumorigenesis. In this study, the impact of CGRP signaling on tumor growth and the associated immune response was investigated using a mouse model of oral cancer.

Lymph nodes are innervated by a unique population of sensory neurons with immunomodulatory potential

Siyi Huang et al.

Summary: The study found that peptidergic nociceptors, a subset of sensory neurons, innervate LNs, distinct from other sensory neurons surrounding the skin. Optogenetic stimulation of LN-innervating sensory fibers triggers rapid transcriptional changes in interacting cell types, particularly endothelium, stromal cells, and innate leukocytes.

FcεR1-expressing nociceptors trigger allergic airway inflammation

Theo Crosson et al.

Summary: Research identified that lung nociceptor neurons amplify immune cell activity and mucus metaplasia in response to an inhaled allergen challenge. By investigating the direct response of neurons to allergens and the effects of nociceptor-specific Fc epsilon R1 gamma knockdown, a feedforward inflammatory loop was revealed in allergic inflammation triggered by allergen sensitization.

STING controls nociception via type I interferon signalling in sensory neurons

Christopher R. Donnelly et al.

Summary: STING is a critical immune regulator that induces type-I interferons and other cytokines to promote immune-cell-mediated clearance of pathogens and neoplastic cells. STING also plays a role in antitumour immunity and has potential as a target for cancer immunotherapy. Additionally, STING is shown to regulate nociception through IFN-I signaling, leading to pain relief in mice and non-human primates.

STING suppresses bone cancer pain via immune and neuronal modulation

Kaiyuan Wang et al.

Summary: STING agonists provide remarkable protection against cancer pain, bone destruction, and local tumor burden through direct neuronal modulation, modulation of immune cell function, and osteoclast function in the tumor microenvironment, offering long-term cancer pain relief.
