Summary: The study found that sinomenine (SIN) regulates the expression of mPGES-1 through its DNA methylation induction effect. The demethylating agent 5-azacytidine (5-AzaC) can reverse the inhibitory effect of SIN on mPGES-1. Additionally, SIN selectively increases the methylation level of the promoter region in the mPGES-1 gene, whereas pretreatment with 5-AzaC suppresses this effect. These findings contribute to predicting and diagnosing diseases with high expression of mPGES-1 and provide new ideas and solutions for clinical diagnosis and treatment as well as drug development.
