The Inhibition of RNA Viruses by Amaryllidaceae Alkaloids: Opportunities for the Development of Broad-Spectrum Anti-Coronavirus Drugs

Shen Tan et al.

Summary: The global COVID-19 pandemic has had a devastating impact on society and vaccines are currently the main medical solution. However, the emergence of vaccine-resistant variants is deemed likely. Therefore, the design and development of broad-spectrum antiviral drugs based on small molecules is seen as a potential alternative strategy.

Biosynthesis of alkaloids in Amaryllidaceae plants: a review

Isabel Desgagne-Penix

Summary: Amaryllidaceae alkaloids are a group of specialized metabolites primarily found in the Amaryllidaceae plant family, with over 600 naturally occurring varieties identified, including galanthamine used to treat neurodegenerative disorders like Alzheimer's disease. Limited commercial availability hinders their clinical development despite promising pharmacological potential. There have been significant advancements in understanding the biosynthesis of Amaryllidaceae alkaloids in recent years through comparative transcriptomic and biochemical approaches.

Flavivirus: From Structure to Therapeutics Development

Rong Zhao et al.

Summary: Flaviviruses pose a hidden threat to global human safety, with urgent need for the development of therapeutic drugs or vaccines. The genome of flavivirus encodes ten proteins, each playing a different and crucial role in viral infection. While significant progress has been made in the understanding of flavivirus biology in recent years, there is currently a lack of fully effective antiviral drugs or vaccines for most flaviviruses.

Lycorine, a non-nucleoside RNA dependent RNA polymerase inhibitor, as potential treatment for emerging coronavirus infections

Young-Hee Jin et al.

Summary: Lycorine has been shown to efficiently inhibit various coronavirus infections, especially MERS-CoV and SARS-CoV-2, by directly inhibiting viral RdRp activity. Its binding affinities with SARS-CoV-2 RdRp residues are higher than those of remdesivir, indicating its potential as a candidate for combating the COVID-19 pandemic.

Amaryllidaceae Alkaloid Cherylline Inhibits the Replication of Dengue and Zika Viruses

Seydou Ka et al.

Summary: The study found that Amaryllidaceae alkaloids (AAs) from plants of the Amaryllidaceae family have strong antiviral activity, with cherylline specifically inhibiting dengue virus and Zika virus replication. This suggests that AAs, including cherylline, could be a potential source of new antiviral drugs.

Amaryllidaceae Alkaloids of Norbelladine-Type as Inspiration for Development of Highly Selective Butyrylcholinesterase Inhibitors: Synthesis, Biological Activity Evaluation, and Docking Studies

Abdullah Al Mamun et al.

Summary: Alzheimer's disease is currently treated with cholinesterase inhibitors and memantine, but targeting butyrylcholinesterase (BuChE) has shown promise in advanced stages of the disease. A series of compounds were synthesized based on Amaryllidaceae alkaloids, showing strong inhibition of human BuChE with good selectivity over AChE. Further studies are needed to explore the potential of these compounds for Alzheimer's disease treatment.
