More than a Catharanthus plant: A multicellular and pluri-organelle alkaloid-producing factory

Summary: Plants are a rich source of natural products, including the monoterpene indole alkaloids (MIAs) which have significant therapeutic potential. However, the supply of MIA drugs has been challenging due to their low yield in plants. This article discusses the recent advances in understanding the biosynthetic pathway and spatial organization of MIAs in plants, which could provide valuable insights for developing metabolic engineering strategies to sustain the MIA supply.

Differential regulation of fluorescent alkaloid metabolism between idioblast and lacticifer cells during leaf development in Catharanthus roseus seedlings

Summary: This study compared the changes in idioblast cells and laticifer cells during Catharanthus roseus seedling growth, and found that the accumulation of fluorescent metabolites in these cells was differentially regulated. Analysis revealed that the fluorescence observed in these cells was emitted from the compound serpentine. The serpentine content in leaves also increased with leaf growth. These findings suggest that idioblast cells and laticifer cells have different biological roles in MIA biosynthesis and regulation.

Engineering Saccharomyces cerevisiae for production of the capsaicinoid nonivamide

Summary: In this study, baker's yeast was engineered to produce nonivamide, a model capsaicinoid, by expressing N-acyltransferases and CoA-ligases of plant and bacterial origin. The constructed yeast platform demonstrated the ability to biosynthesize nonivamide for the first time, showing potential for in vivo biocatalytic formation of capsaicinoids and as a tool for the discovery of novel drugs.

The terminal enzymatic step in piperine biosynthesis is co-localized with the product piperine in specialized cells of black pepper (Piper nigrum L.)

Summary: This study provides experimental evidence on the cellular localization and biosynthesis of piperine and piperamide synthase in specific cells of black pepper fruits. Piperine and piperamide synthase accumulate during early stages of fruit development and decline before maturity, and their co-localization is evidenced by fluorescence microscopy. The increasing levels of piperine during fruit maturation are consistent with the increasing numbers of fluorescent cells within the fruit perisperm.

The results demonstrate differential sensitivity of CM334 and Dempsey to Agrobacterium-mediated callus induction, as well as differential selection pressure of PPT through the pBAtC binary vector.

Some extremely pungent varieties show similarities in morphology between the pericarp and placental septum. These findings contribute to a better understanding of capsaicinoid production in chili pepper fruits.
