Summary: Although strain SDTB022 showed poor direct inhibitory effect against the mycelial growth of Phytophthora parasitica var. nicotianae, it promoted plant growth, increased yield, and enhanced plant defense responses. Through the upregulation of defense enzymes and systemic acquired resistance pathways in tobacco, B. velezensis SDTB022 has the potential to effectively control TBS disease and could be used as a biological pesticide in modern agriculture.

Efficient production of extracellular alkaline protease in Bacillus amyloliquefaciens by host strain construction

Summary: In this study, a high extracellular protein expression capacity host of Bacillus amyloliquefaciens, named BAX-10, was developed by deleting seven important extracellular protease genes, one important intracellular protease gene, and two redundant protein genes. The alkaline protease gene aprE from Bacillus subtilis D7 was expressed in BAX-10, resulting in higher alkaline protease activity compared to the control strain. Furthermore, the deletion of the ten genes did not negatively affect the growth of B. amyloliquefaciens. Therefore, B. amyloliquefaciens BAX-10 could be a potential platform host for expressing target extracellular proteins.

Comparative genomics analysis of Bacillus velezensis LOH112 isolated from a nonagenarian provides insights into its biocontrol and probiotic traits

Summary: This study evaluated the beneficial traits of a newly isolated B. velezensis strain LOH112 using comprehensive bioinformatics and comparative genomic analyses and in vitro experimental approaches. The results showed that LOH112 has broad spectrum antibacterial and antifungal activities, hydrolyzes protein and cellulose, and forms strong adhesive biofilms. LOH112 contains multiple secondary metabolism gene clusters responsible for antimicrobial compound production, as well as strain-specific orthologous gene clusters related to proteomic homeostasis. These findings suggest the potential applications of LOH112 as biocontrol and probiotic agents.

Evaluating Enzymatic Productivity-The Missing Link to Enzyme Utility

Summary: Kinetic productivity analysis is crucial for characterizing enzyme catalytic performance and capacity, but it is often overlooked in published literature. Only a small fraction of studies provide productivity analysis, despite it being the only reliable indicator of potential commercial value. This article argues for the importance of reporting productivity data for native, modified, and immobilized enzymes under different reaction conditions to optimize enzymatic processes and accelerate biotechnological applications. Using examples from various studies, the article demonstrates the critical significance of productivity in the translational and commercial use of enzymes and the processes employing them. The article concludes by suggesting steps to maximize enzyme catalyzed reaction productivity.

The antioxidant peptides were obtained through protein hydrolysis and microbial fermentation. The purified peptides showed high oxygen radical absorbance capacity (ORAC) and their scavenging activity depended on the content and position of certain amino acids in the sequences. LC-MS/MS analysis identified several peptides with antioxidant properties.

Summary: Chronic oxidative stress plays a key role in beta-cell loss in diabetes, and recent studies have linked this loss to ferroptotic-like death. Oxidative stress leads to the loss of reduced/oxidized glutathione and degradation of glutathione peroxidase 4, promoting lipid peroxidation and ferroptotic-like cell damage. Inhibiting the c-Abl kinase and inducing ERRy preserves beta-cell function by increasing the GSH/GSSG ratio and inhibiting lipid peroxidation. This study provides evidence that ERRy-induced GLS1 expression augments the glutathione antioxidant system, improving beta-cell function and survival under oxidative stress conditions.
