Increasing fengycin production by strengthening the fatty acid synthesis pathway and optimizing fermentation conditions

Summary: Bacillus subtilis 168 was engineered as a cell factory for fengycin production by activating its dormant native fengycin synthetase. Genetically modified strain 168DS showed an initial fengycin production of 10.40 mg/L in flask culture, which increased to 130.10 mg/L through a combination of gene overexpression and optimization of fermentation conditions. This strategy provides a path to increase industrial production and reduce the price of fengycin.

Relationship between lipopeptide biosurfactant and primary metabolite production by Bacillus strains in solid-state and submerged fermentation

Purification and characterization of antibacterial surfactin isoforms produced by Bacillus velezensis SK

Summary: Bacillus velezensis SK, isolated from soil, exhibits broad-spectrum antimicrobial activity. Efficient extraction of antimicrobial compounds produced in different mediums was achieved using Diaion HP-20 resin. The antimicrobial compound, characterized by TLC, FTIR, and in-situ bioautography analysis, was found to be cyclic lipopeptides and was purified using silica gel, size exclusion, dual gradient, and RP-HPLC chromatography techniques. Growth kinetic studies showed that Bacillus velezensis SK produces a mixture of lipopeptides. LC-ESI-MS analysis of partially purified lipopeptide identified surfactin variants, and further analysis revealed the presence of C-15-surfactin homologues. The isolated surfactin exhibited good antimicrobial activity against drug-resistant bacteria.

LPs have broad-spectrum antimicrobial activities and can effectively inhibit microbial spoilage of food, playing an important role in preserving the quality of vegetables, fruits, drinks, and aquatic products. In addition, the yield of LPs can be enhanced by gene regulation and the optimization of fermentation parameters, and nano-encapsulation of LPs holds promise for improving the preservation of food.
