Synthesis of Fluorinated Amides Starting from Carbohydrates Based on the Claisen Rearrangement

Monika Bilska-Markowska et al.

Summary: This study presents an efficient synthetic protocol for the synthesis of fluorine-containing amides derived from sugar, utilizing the Claisen rearrangement of previously obtained fluorinated vinyl ethers. The newly synthesized compounds were characterized using NMR spectroscopy, mass spectrometry, and X-ray crystallography. The resulting molecules, with potential for further functional group transformation, can serve as valuable fluorinated scaffolds for the synthesis of subsequent chemical compounds.

Deoxyfluorinating reagents as tools for γ-amino-α-hydroxyphosphonate modification

Marcin Kazmierczak et al.

Summary: This article introduces the application of deoxyfluorinating reagents DAST and PyFluor, which can be used for selective modification of gamma-amino-alpha-hydroxyphosphonates. Depending on the specific deoxyfluorinating reagents used, intramolecular cyclization or direct nucleophilic deoxyfluorination can be observed, leading to different derivatives. These compounds can serve as precursors for important medicinal compounds.

Tetraalkyl Hydroxymethylene-bisphosphonate and Dialkyl 1-Diphenylphosphinoyl-1-hydroxy-ethylphosphonate Derivatives by the Pudovik Reaction and Their Rearranged Products

Zsuzsanna Szalai et al.

Summary: The reaction of diethyl alpha-oxoethylphosphonate and diethyl oxobenzylphosphonate with various phosphites and phosphine oxides can lead to the formation of Pudovik adducts and/or rearranged products, depending on the substrates and reaction conditions. The nature of the substituent on the central carbon atom influences the tendency for rearrangement, and asymmetric products with different functionalities show interesting NMR features.

Probing cytochrome P450 (CYP) bioactivation with chloromethylindoline bioprecursors derived from the duocarmycin family of compounds

Natalia Ortuzar et al.

Summary: This study examines the potential cytochrome P450 (CYP)-mediated cancer cell killing ability of seco-OH-chloromethylindoline (CI) duocarmycin-based bioprecursors. Results demonstrate up to a 10-fold increase in bioactivation of de-OH CI-MI and a fluoro bioprecursor analogue in CYP1A1-transfected cells. Additionally, it is shown that CYP1A2, but not CYP1B1 or CYP3A4, has a propensity for potentiating these compounds, indicating a preference for CYP1A bioactivation.
