Jolkinolide B sensitizes bladder cancer to mTOR inhibitors via dual inhibition of Akt signaling and autophagy

Jun Sang et al.

Summary: This study found that the combination therapy of a natural anti-cancer agent Jolkinolide B (JB) and mTOR inhibitors (mTORi) can effectively treat bladder cancer. JB inhibits Akt signaling and cytoprotective autophagy, enhancing the anti-proliferative effects of mTORi. Furthermore, mTORi enhances the pro-apoptotic and pro-paraptotic effects of JB through activating endoplasmic reticulum stress and MAPK pathways.

Advances on Natural Abietane, Labdane and Clerodane Diterpenes as Anti-Cancer Agents: Sources and Mechanisms of Action

Rosaria Acquaviva et al.

Summary: Extensive research has identified numerous phytochemicals as potential sources of anti-cancer compounds. Natural compounds are considered suitable candidates for the development of new anti-cancer drugs due to their pleiotropic actions. This comprehensive review focuses on the screening of phytochemicals, particularly diterpenes with abietane, clerodane, and labdane skeleton, for anticancer drug development, discussing their chemical structure, natural sources, and mechanisms of action.

Sugiol, a diterpenoid: Therapeutic actions and molecular pathways involved

Vivek K. Bajpai et al.

Summary: Understanding the interaction between natural product structural diversity, cellular metabolism, and infectious disease targets remains challenging. Inflammation plays a crucial role in the human healing process, and drugs derived from plant products, including sugiol, are being actively researched. Despite evidence of efficacy and safety, sugiol has not been approved as a therapeutic agent due to low bioavailability and insolubility in an aqueous environment.
