MAP7D2 reduces CD8+cytotoxic T lymphocyte infiltration through MYH9-HMGB1 axis in colorectal cancer

Summary: The study showed that neoadjuvant toripalimab with or without celecoxib could be a potential therapeutic option for patients with mismatch repair-deficient or microsatellite instability-high, locally advanced colorectal cancer. This treatment was associated with a high pathological complete response rate and an acceptable safety profile, which did not compromise surgery. Longer term follow-up is needed to assess effects on survival-related endpoints.

Extracellular HMGB1 blockade inhibits tumor growth through profoundly remodeling immune microenvironment and enhances checkpoint inhibitor-based immunotherapy

Summary: In this study, it was demonstrated that targeting extracellular HMGB1 can significantly remodel the tumor immune microenvironment for more effective cancer therapy. Inhibition of HMGB1 led to reductions in immunosuppressive cells, such as myeloid-derived suppressor cells and regulatory T lymphocytes, while also enhancing the activation of immune cells like dendritic cells. Additionally, blocking HMGB1 improved the efficacy of anti-PD-1 cancer immunotherapy, suggesting that extracellular HMGB1 blockade could enhance current cancer immunotherapies.

Summary: This study reveals a cell-extrinsic role of KRAS in modulating the tumor microenvironment by reprogramming tumor-associated macrophages, promoting tumor progression and inducing resistance to cetuximab therapy. Mechanistically, mutant KRAS stabilizes HIF-1 alpha to drive the production of CSF2 and lactate in tumor cells, enhancing HIF-1 alpha stabilization and contributing to a permissive microenvironment for tumor malignancy.
