Exosome-liposome hybrid nanoparticle codelivery of TP and miR497 conspicuously overcomes chemoresistant ovarian cancer

Longxia Li et al.

Summary: This study investigated the combined application of miR497 and TP in overcoming cisplatin-resistant ovarian cancer (OC) by suppressing the mTOR signaling pathway. Bioinspired hybrid nanoparticles were prepared to deliver miR497 and TP, showing efficient uptake by tumor cells and significant anticancer activity in vitro and in vivo. Mechanistically, the nanoparticles promoted dephosphorylation of the overactivated PI3K/AKT/mTOR signaling pathway and enhanced reactive oxygen species (ROS) generation and macrophage polarization. Overall, this study offers a translational strategy to overcome cisplatin-resistant OC and potentially treat other cisplatin-resistant tumors.

Summary: Multiple myeloma is a malignancy characterized by plasma cell accumulation in the bone marrow. Within the tumor microenvironment, macrophages in multiple myeloma exhibit pro-myeloma functions, which are modulated by the polycomb-group protein BMI1. Knockout of BMI1 in MM-associated macrophages leads to reduced proliferation, decreased expression of angiogenic factors, and loss of protection against chemotherapy-induced cell death, ultimately negating the pro-myeloma effects of these cells.
