LCAT- targeted therapies: Progress, failures and future

Kaixu Yang et al.

Summary: This article mainly discusses the role of lecithin: cholesterol acyltransferase (LCAT) in high-density lipoprotein metabolism and reverse cholesterol transport, as well as the diseases and therapies associated with LCAT. Increasing LCAT activity to improve plasma lipid metabolism has become a research focus, with potential as an effective treatment for familial LCAT deficiency, fish eye disease, and cardiovascular disease.

Summary: LXR agonists have potential anti-atherosclerotic effects by increasing cholesterol efflux, but may cause liver toxicity and hypertriglyceridemia. Encapsulating the LXR agonist in synthetic HDL nanoparticles improves drug efficacy and reduces side effects.

Apolipoprotein Mimetic Peptides: Potential New Therapies for Cardiovascular Diseases

Anna Wolska et al.

Summary: Since the breakthrough of using insulin to treat diabetic patients in the 1920s, there has been a growing interest in developing various proteins and their peptide mimetics for treating medical disorders. Currently, there are over 60 approved peptides for human use and more than 150 peptides in clinical development. This review mainly focuses on peptide mimetics related to cardiovascular diseases, discussing the biochemistry, design, and clinical trials of peptides based on apoA-I, apoE, and apoC-II, while also addressing the limitations and challenges in developing apolipoprotein mimetic peptides into new drugs.

Temperature- and composition-dependent conformational transitions of amphipathic peptide-phospholipid nanodiscs

Chiharu Anada et al.

Summary: The study explored the temperature- and composition-dependent structural changes of 18A-phosphatidylcholine complexes. It was found that above the gel-to-liquid-crystalline phase transition temperature, nanodiscs increased in size, forming enlarged nanodiscs and a lamellar phase. Further increase in temperature induced the formation of lipid vesicles. These transformations were explained using a transition model based on the migration of the peptide from the rim of the nanodiscs to the liquid-crystalline bilayer phase.

High-density lipoproteins, reverse cholesterol transport and atherogenesis

Henry J. Pownall et al.

Summary: Plasma HDL-cholesterol levels are negatively correlated with the risk of ASCVD, but genetic approaches to increase HDL levels do not reduce this risk. The mechanism of reverse cholesterol transport involving FCh transfer between HDL and macrophages is still under debate in relation to ASCVD.

Positive allosteric modulators of lecithin: Cholesterol acyltransferase adjust the orientation of the membrane-binding domain and alter its spatial free energy profile

Akseli Niemela et al.

Summary: This study utilized atomistic molecular dynamics simulations and free energy calculations to investigate the effects of a set of positive allosteric modulators on the structural and dynamical properties of LCAT. The results suggest that these compounds may promote the activity of LCAT by inducing a different spatial orientation and sampling of the membrane-binding domain (MBD), facilitating the lid's movement from closed to open state.

LCAT deficiency: a systematic review with the clinical and genetic description of Mexican kindred

Roopa Mehta et al.

Summary: This systematic review shows that LCAT deficiency syndromes are clinically and genetically heterogeneous. There was a significantly greater risk of premature coronary artery disease in fish eye disease compared to familial LCAT deficiency. The LCAT mutations discussed in this article are the only ones reported in the Mexican-Amerindian population.
