Monitoring the ecological status of rivers with diatom eDNA metabarcoding: A comparison of taxonomic markers and analytical approaches for the inference of a molecular diatom index

Laure Apotheloz-Perret-Gentil et al.

Summary: Recent studies have shown the usefulness of diatom eDNA metabarcoding for assessing river and stream ecological quality. However, differences in taxonomic markers and data analysis methods hinder result comparison. This study compared two taxonomic markers and three analytical approaches and found that taxonomy-free methods were more correlated with morphological index values, with rbcL marker showing higher correlations than 18S marker.

The potential of exact sequence variants (ESVs) to interpret and assess the impact of agricultural pressure on stream diatom assemblages revealed by DNA metabarcoding

Kalman Tapolczai et al.

Summary: This study assessed the impact of land-use on diatom assemblages using DNA metabarcoding and tested a taxonomy-free approach for a better interpretation of agricultural stress. Results showed that the taxonomy-free approach provided better insights into the effects of agricultural land use and could lead to the development of more efficient indices. Furthermore, it demonstrated the importance of retaining ecological information by using exact sequence variants without taxonomic assignment.

DNA metabarcoding reveals differences in distribution patterns and ecological preferences among genetic variants within some key freshwater diatom species

Javier Perez-Burillo et al.

Summary: This study evaluates the genetic variants within several ecologically important diatom species and finds that different genetic variants within the same species show diverse distribution patterns and ecological preferences, with varying responses to environmental variables. The results demonstrate the importance of studying genetic variants within species complexes for understanding ecological tolerance and developing accurate biological indexes for biomonitoring programs.

Cyprus' diatom diversity and the association of environmental and anthropogenic influences for ecological assessment of rivers using DNA metabarcoding

Panayiota Pissaridou et al.

Summary: Human activities are the leading cause of environmental impairments. A study in Cyprus found differences in diatom assemblages between intermittent and perennial rivers, with Achnanthidium minutissimum being more abundant in intermittent rivers and Amphora pediculus and Planothidium caputium in perennial ones. Factors such as nutrients, station characteristics, and land use activities have an impact on the observed differences in diatom diversity.

Summary: The study showed that excluding diatom species primarily driven by mass effect from bioassessment leads to a more accurate determination of river ecological status through the modified Biological Diatom Index (IBDmod), especially in conditions of moderate nutrient enrichment.
