Neoadjuvant Chemotherapy Induces Genomic and Transcriptomic Changes in Ovarian Cancer

Melissa Javellana et al.

Summary: This study investigated the changes in the genome and transcriptome of patients with advanced high-grade serous ovarian cancer (HGSOC) receiving neoadjuvant chemotherapy. The results showed increased AP-1 activity and SIK2 copy-number amplifications in resistant ovarian cancer following chemotherapy, and demonstrated synergistic effects of AP-1 and SIK2 inhibitors with chemotherapy.

BMP2-induction of FN14 promotes protumorigenic signaling in gynecologic cancer cells

Tomohiko Fukuda et al.

Summary: BMP2 induces FN14 expression, promoting proliferation and migration of ovarian and endometrial cancer cells, as well as modulating the expression of c-KIT and SNAIL, which are crucial for cancer cell growth and migration.

Characterization of SOX2, OCT4 and NANOG in Ovarian Cancer Tumor-Initiating Cells

Summary: The findings suggest that SOX2 may be a more consistent indicator of ovarian tumor initiating cells (TICs) that contribute to tumor repopulation following chemotherapy. Further studies evaluating SOX2 in TIC biology will enhance understanding of mechanisms driving ovarian cancer relapse.

Cancer-associated mesothelial cells promote ovarian cancer chemoresistance through paracrine

Jin Qian et al.

Summary: Ovarian cancer-associated mesothelial cells play a critical role in promoting chemoresistance and stemness in ovarian cancer. The secreted factor osteopontin drives mesothelial-mediated chemoresistance and stemness, and therapeutic targeting of osteopontin may enhance platinum sensitivity in ovarian cancer.

Small Molecule Targeting of Oxysterol-Binding Protein (OSBP)-Related Protein 4 and OSBP Inhibits Ovarian Cancer Cell Proliferation in Monolayer and Spheroid Cell Models

Ryan C. Bensen et al.

Summary: ORP4 is identified as a potential druggable target in ovarian cancer cells, and the natural product compound OSW-1 exhibits high antiproliferative potency by targeting ORP4 and OSBP. The absence of extracellular lipids significantly enhances the cytotoxicity of OSW-1, suggesting a potential synergistic effect in the in vivo ovarian cancer microenvironment when OSBP is also targeted.

Frizzled-7 Identifies Platinum-Tolerant Ovarian Cancer Cells Susceptible to Ferroptosis

Yinu Wang et al.

Summary: The study identifies platinum-tolerant cancer cells with cancer stem cell features characterized by expression of FZD7 and dependency on the FZD7-beta-catenin-Tp63-GPX4 pathway for survival. These cells are highly susceptible to ferroptosis, marking a novel therapeutic vulnerability in platinum-resistant cancer cells.
