Drug-dependent inhibition of nucleotide hydrolysis in the heterodimeric ABC multidrug transporter PatAB from Streptococcus pneumoniae

Summary: In this study, the authors used mass spectrometry techniques to determine the subunit stoichiometry and investigate drug binding locations of the multidrug resistance protein PatAB. The results suggest that the drug ethidium binds to the nucleotide-binding domain of PatA and can regulate nucleotide hydrolysis. Additionally, other drugs such as propidium, novobiocin, and coumermycin A1 were found to inhibit nucleotide hydrolysis by a non-competitive mechanism.

Summary: Macrolides such as clarithromycin have been shown to inhibit pneumolysin release in macrolide-resistant Streptococcus pneumoniae (MRSP). Clarithromycin downregulates the gene encoding pneumolysin, decreases pneumococcal growth, and improves lung injury in a mouse model of pneumonia. Targeting pneumolysin may be a promising strategy for treating pneumococcal pneumonia.
