Summary: The study successfully delivered mRNA or single-guide RNA for gene editing in vivo using combinatorially synthesized ionizable phospholipids. Optimized iPhos lipids can change shapes in endosomal acidic environments to facilitate the release of gene editing materials. When working synergistically with helper lipids, iPhos lipids can achieve targeted delivery to specific organs.

Lumasiran, an RNAi Therapeutic for Primary Hyperoxaluria Type 1

Summary: Primary hyperoxaluria type 1 is a rare genetic disease caused by hepatic overproduction of oxalate, leading to kidney stones, nephrocalcinosis, kidney failure, and systemic oxalosis. This trial tested the efficacy of lumasiran, an RNAi therapeutic agent, in reducing hepatic oxalate production. The results showed that lumasiran significantly reduced urinary oxalate excretion, alleviating the cause of kidney failure in PH1 patients.

Optimization of PEGylated KL4 Peptide for siRNA Delivery with Improved Pulmonary Tolerance

Summary: Covalent attachment of monodisperse poly(ethylene glycol) (PEG) to KL4 peptide improves its solubility and enhances uptake into lung epithelial cells, leading to robust gene silencing effects. Among PEGylated KL4 peptides, PEG(12)KL4 was found to be optimal for siRNA delivery with low risk of inflammatory response and toxicity.
