Assessing and counteracting fibrosis is a cornerstone of the treatment of CKD secondary to systemic and renal limited autoimmune disorders

Marc Xipell et al.

Summary: Chronic kidney disease (CKD) is a growing global cause of morbidity and mortality. Immune mediated disorders and the dysregulation of extracellular matrix synthesis and degradation play crucial roles in the development of CKD and renal fibrosis. Pathways involved in renal fibrosis and CKD are closely related to each other, and their understanding and translation into clinical practice are still necessary. The future holds promise for more personalized assistance for CKD and renal fibrosis with the rapid development of technology and the availability of new drugs in nephrology.

Inhibition of the C3a receptor attenuates sepsis-induced acute lung injury by suppressing pyroptosis of the pulmonary vascular endothelial cells

Zhan-Fei Li et al.

Summary: Acute lung injury (ALI) is a leading cause of bacterial sepsis-related death. Studies have shown that C3a levels are significantly increased in organs affected by sepsis and are closely associated with organ dysfunction and poor prognosis. This study found that C3a plays an important role in preventing sepsis-induced lung endothelial cell (EC) pyroptosis.

The IL-18/IL-18R1 signalling axis: Diagnostic and therapeutic potential in hypertension and chronic kidney disease

Jordyn M. Thomas et al.

Summary: Chronic kidney disease (CKD) is an inflammatory condition, and the inflammasome and pro-inflammatory cytokine IL-18 play important roles in its development. Regulating the signaling pathways of the inflammasome and IL-18 may serve as potential targets for CKD treatment.

Redox signaling pathways in unilateral ureteral obstruction (UUO)-induced renal fibrosis

Ana Karina Aranda-Rivera et al.

Summary: In the UUO model, overproduction of ROS leads to alterations in signaling pathways, promoting inflammation, oxidative stress, and apoptosis, contributing to fibrosis development. Impairment of mitochondrial metabolism is also involved in this process.

Liproxstatin-1 attenuates unilateral ureteral obstruction-induced renal fibrosis by inhibiting renal tubular epithelial cells ferroptosis

Bo Zhang et al.

Summary: The study demonstrates the role of ferroptosis in renal fibrosis induced by unilateral ureteral obstruction. Inhibiting ferroptosis can alleviate the morphological and pathological changes of renal fibrosis and suppress the expression of profibrotic factors. Liproxstatin-1 may potentially be used as a therapeutic approach for UUO-induced renal fibrosis by inhibiting ferroptosis in renal tubular epithelial cells.
