Unique molecular and functional features of extramedullary hematopoietic stem and progenitor cell reservoirs in humans

Nicole Mende et al.

Summary: By using single-cell RNA-sequencing and functional assays, we comprehensively delineate the characteristics of extramedullary HSPC compartments in humans. We found shared features of HSPCs in spleen, peripheral blood, and mobilized peripheral blood, as well as a unique bias towards erythroid-megakaryocytic differentiation in healthy peripheral blood HSPCs. Our study provides important insights into the process of blood production and the potential clinical diagnosis and monitoring.

Sequential Stem Cell-Kidney Transplantation in Schimke Immuno-osseous Dysplasia

Alice Bertaina et al.

Summary: This study aimed to find a new approach to eliminate immune rejection and the need for post-transplantation immunosuppression. By performing bone marrow and kidney transplantation on three patients with a specific genetic condition, the researchers successfully achieved the goal of normal renal function without the need for immunosuppression.

Transplantation Tolerance through Hematopoietic Chimerism

Thomas R. Spitzer et al.

Summary: The advances in human transplantation have been hindered by the immunologic consequences of histoincompatibility between donor and recipient. Finding solutions to achieve successful hematopoietic stem-cell and solid-organ transplantation without immune complications is the ultimate goal in medical science.

Lymphohematopoietic graft-versus-host responses promote mixed chimerism in patients receiving intestinal transplantation

Jianing Fu et al.

Summary: In patients receiving intestinal transplants, long-term multilineage blood chimerism often develops, especially with donor T cell macrochimerism. These macrochimeras tend to have high GvH to HvG T cell clonal ratios in their allografts, suggesting a potential role in reducing rejection without causing GVHD. The migration of tissue-resident GvH-reactive donor T cells into the recipient circulation and bone marrow may contribute to hematopoietic cell destruction and the engraftment of graft-derived HSPCs to maintain chimerism.

Detection of Hematopoietic Stem Cell Transcriptome in Human Fetal Kidneys and Kidney Organoids Derived From Human Induced Pluripotent Stem Cells

Jin Wook Hwang et al.

Summary: Cells expressing HSC transcriptome were found in the human fetal kidney, and similar expression features were observed in iPSC-derived kidney organoids. These findings support the presence of HSC markers in the human kidney. Further investigation is needed to understand the mechanisms behind the appearance of these cells with similar transcriptional features.
