RNA Granules in Antiviral Innate Immunity: A Kaposi's Sarcoma-Associated Herpesvirus Journey

Nishi R. Sharma et al.

Summary: RNA granules are important for post-transcriptional gene regulation. KSHV, a DNA virus, counteracts RNA granules through the expression of ORF57 protein, allowing it to evade host antiviral immunity and ensure productive infection.

Shiftless, a Critical Piece of the Innate Immune Response to Viral Infection

William Rodriguez et al.

Summary: SHFL, as an interferon-stimulated gene, plays a critical role in the innate immune response to viral infection by restricting viral replication through diverse mechanisms across different types of viruses. It not only affects viral RNA stability and translation, but also directly inhibits specific translation strategies utilized by some viruses.

Regulation of processing bodies: From viruses to cancer epigenetic machinery

Sunmathy Kanakamani et al.

Summary: Processing bodies (PBs) are cytoplasmic messenger ribonucleoprotein particle granules involved in eukaryotic gene expression regulation. They are maintained by RNA-protein and protein-protein interactions and undergo dynamic changes in response to cellular cues, including viral infections. Some PB proteins are implicated in cancer development, but the effects of epigenetic modifications on cancer-associated PB genes on PB dynamics remain to be fully explored.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Shiftless inhibits flavivirus replication in vitro and is neuroprotective in a mouse model of Zika virus pathogenesis

Natasha W. Hanners et al.

Summary: Flaviviruses like Zika virus and West Nile virus can cause severe neuropathology if they invade the central nervous system. The interferon-stimulated gene (ISG) effector Shiftless (SHFL) has been identified as a potent inhibitor of various positive-stranded RNA viruses, including multiple Flaviviridae members, and plays an important role in host protection against Zika virus infection and virus-induced neuropathological disease.

EGR1 Suppresses Porcine Epidemic Diarrhea Virus Replication by Regulating IRAV To Degrade Viral Nucleocapsid Protein

Hua Wang et al.

Summary: PEDV is a globally distributed alphacoronavirus that has reemerged recently, causing significant economic losses. This study found that PEDV infection induced the expression of EGR1 in PEDV-permissive cells, which suppressed viral replication by upregulating IRAV expression. EGR1-mediated viral restriction provides a potential therapeutic target for controlling PEDV infection.

Viral Manipulation of a Mechanoresponsive Signaling Axis Disassembles Processing Bodies

Elizabeth L. Castle et al.

Summary: The viral protein KapB from Kaposi's sarcoma-associated herpesvirus induces PB disassembly by promoting actin rearrangements, RhoA effectors, and the mechanoresponsive transcription activator YAP. Ectopic expression of active YAP or exposure of ECs to mechanical forces can also cause PB disassembly in the absence of KapB. This study suggests that KapB activates a mechanoresponsive signaling axis to enhance inflammatory molecule expression through changes in cell shape and cytoskeletal structures.
