Detection of the ABCB11930_1931del TC Mutation in Two Suspected Ivermectin-Sensitive Cats and Their Relatives by a Novel TaqMan Allelic Discrimination Assay

Daniela Nuernberger et al.

Summary: This study supports previous findings on drug sensitivity in cats with the homozygous ABCB11930_1931del TC mutation. The newly established TaqMan allelic discrimination assay provides a useful and reliable method for routine MDR1 genotyping in cats in order to identify drug-sensitive cats prior to treatment with established P-gp substrates such as ivermectin and other macrocyclic lactones and thus improve therapeutic safety.

Clinical Relevance of Hepatic and Renal P-gp/BCRP Inhibition of Drugs: An International Transporter Consortium Perspective

Kunal S. Taskar et al.

Summary: This article discusses the role of P-glycoprotein (P-gp) and breast cancer resistance protein (BCRP) in drug-drug interactions, drug absorption, and brain penetration. Based on clinical evidence, the article suggests that inhibition of P-gp or BCRP in the liver or kidneys has limited clinical implications on drug disposition.

Role of an ABCB11930_1931del TC gene mutation in a temporal cluster of macrocyclic lactone-induced neurologic toxicosis in cats associated with products labeled for companion animal use

Katrina L. Mealey et al.

Summary: The study found a potential link between the ABCB11930_1931del TC gene mutation and neurologic toxicosis in cats exposed to eprinomectin-containing antiparasitic products. This mutation appears to be rare in the general cat population, but veterinarians should be aware of its existence and potential impact on enhancing susceptibility to adverse drug reactions.

Tolerance and Pharmacokinetics of Galliprant™ Administered Orally to Collies Homozygous for MDR1-1Δ

Mark C. Heit et al.

Summary: The study aimed to evaluate the pharmacokinetics and tolerance of grapiprant in collies homozygous for MDR1-1 Delta. It found that the drug was well tolerated in these dogs with increased exposure, except for self-limiting vomiting as a treatment-related clinical sign. Population pharmacokinetic analysis suggested differences in drug handling between these dogs and normal dogs.
