A parameter study of ultrasound assisted enzymatic esterification

Anamaria Vartolomei et al.

Summary: This work focuses on studying the esterification parameters for the ultrasound assisted synthesis of isoamyl acetate using lipase Lipozyme 435 in a continuous loop reactor. The results show that a higher concentration of ester can be achieved at specific conditions, and the best ultrasound conditions are a short continuous sonication time, ultrasound power of 32 mW, and amplitude of 20%. The enzyme can be successfully reused three times without losing its activity, and the reaction kinetics indicate that satisfactory reaction concentrations can be reached in a short time.

Multienzyme System in Amorphous Metal-Organic Frameworks for Intracellular Lactate Detection

Yuanyu Zhang et al.

Summary: This study establishes a multienzyme system consisting of lactate oxidase (LOx) and horseradish peroxidase (HRP) in living cells using amorphous metal-organic frameworks (aMOFs). This system, acting as artificially constructed organelles, shows high activity and selectivity for intracellular lactate detection, significantly improving the signal to noise ratio. The concept of constructing multienzyme systems holds promise for detecting various intracellular metabolites.

Summary: This study focuses on modifying the functionality and ratio of linkers in ZIFs using the MTV approach to optimize the activity of encapsulated enzymes. The research revealed a nonlinear correlation between enzyme activity and the ratio of hydrophilic linkers in MTV-ZIF-8, indicating the possibility of changing enzyme structure through conformation changes. The optimized BCL@ZIF-8 shows improved thermal and chemical stability compared to free BCL in solution, and also doubles the catalytic reactivity in kinetic resolution reactions.
