Lifespan extension with preservation of hippocampal function in aged system xc--deficient male mice

Summary: Deletion of the xCT subunit of system x(c)(-) has been found to extend lifespan and promote healthy aging in mice. Although aged xCT(-/-) mice had higher plasma cystine/cysteine ratio, it did not negatively affect their overall health status. On the contrary, the age-related priming of the innate immune system was attenuated in xCT(-/-) mice, and these mice showed preserved hippocampal function and retention of hippocampus-dependent memory.

Summary: The key event in the pathophysiology of stroke is anoxic depolarization (AD) of neurons in the ischemic core, and the time between AD and re-oxygenation determines the synaptic recovery after anoxic challenge. Adenosine released during anoxia suppresses synaptic glutamate release, while antagonism of glutamate receptors prolongs AD latency and alters depolarizing waves. In experiments, AD latency was longer in CA1 than CA3, with differences between genders.

Summary: The study showed that the expression of xCT gene and protein is upregulated early after spinal cord injury (SCI) and negatively impacts motor neuron survival. Deletion of xCT modulates intraspinal glial activation towards an anti-inflammatory profile, leading to faster recovery of muscular grip strength and pre-SCI weight in xCT-deficient mice.
