Charged groups on pyropheophorbide-based photosensitizers dictate uptake by tumor cells and photodynamic therapy efficacy

Courtney Saenz et al.

Summary: This study investigated the impact of anionic and cationic substituents of the pyropheophorbide-based photosensitizers (PS) on uptake and retention by tumor epithelial cells and photodynamic therapy (PDT). The study found that the nature of the functionalities in the macrocycle made a significant difference in overall lipophilicity and in binding to and retention by tumor cells. Cationic PSs showed higher cellular binding, uptake, and in vitro PDT efficacy than anionic PSs.

Photodynamic therapy for squamous cell carcinoma of the head and neck: narrative review focusing on photosensitizers

Ana Melissa Ccopa Ibarra et al.

Summary: The effectiveness of photodynamic therapy in squamous cell carcinoma of the head and neck varies depending on the type of photosensitizer, tumor location, and stage. While promising for early-stage SCCHN, there is limited evidence supporting its efficacy in advanced tumors. Adverse effects were generally well tolerated by patients, suggesting potential for further clinical trials to determine its full effectiveness.

Factors Affecting Photodynamic Therapy and Anti-Tumor Immune Response

Michael R. Hamblin et al.

Summary: Photodynamic Therapy (PDT) is a cancer treatment that utilizes a photosensitizer to activate reactive oxygen species, killing tumor cells and stimulating the host immune system. Factors such as the structure of the photosensitizer, drug-light interval, rate of light delivery, and combination with adjuvants can affect the efficiency of the immune response. The success of checkpoint inhibitors in tumor immunotherapy opens up the possibility for investigating PDT in combination with other approaches in clinical scenarios.

Characteristics of an Impaired PDT Response

David Kessel et al.

Summary: HPV infection can enhance the efficacy of ionizing radiation for head and neck cancer treatment, while affecting the effectiveness of photodynamic therapy by promoting different cell death pathways. The explanation for these differences in PDT efficacy needs further exploration.

Cell-type deconvolution analysis identifies cancer-associated myofibroblast component as a poor prognostic factor in multiple cancer types

Bingrui Li et al.

Summary: This study comprehensively characterized cell-type alterations across 18 cancer types and identified myofibroblastic CAF (myCAF) composition as a poor prognostic factor in nine cancer types. Furthermore, genes associated with the differentiation degree of myCAF and inflammatory CAF (iCAF) were identified, highlighting their roles in fibroblast and extracellular matrix-related pathways. These findings provide therapeutic perspectives for targeting CAF subtypes in the tumor microenvironment and arranging treatment schemes based on CAF compositions in different cancer types.

3D Cancer Models: Depicting Cellular Crosstalk within the Tumour Microenvironment

Teresa Franchi-Mendes et al.

Summary: The tumour microenvironment consists of multiple non-cancerous cells that interact with tumour cells, influencing disease progression and therapeutic responses. In vitro 3D models have been developed to mimic the complexity of the tumour microenvironment, providing insights for cancer research and drug discovery. Understanding the role of each cell type in the microenvironment is crucial for uncovering potential therapeutic targets.
