Summary: This article describes the synthesis and characterization of a small series of tethered ruthenium(II) eta(6)-arene complexes, where a single benzylic substituent is examined to enforce chirality at the metal center upon ligation of a tethered bidentate ligand. The application of these complexes as catalysts in the asymmetric transfer hydrogenation of ketones is discussed, and moderate enantioselectivities confirm the validity of the approach.

Summary: A practical method for the asymmetric transfer hydrogenation/dynamic kinetic resolution of N-Boc 3-fluoro-dihydrotetrahydroquinolin-4-ones into the corresponding cis-fluoro alcohols is reported. The reaction achieved high yields, up to 99:1 diastereomeric ratio (dr), and up to >99% enantiomeric excess (ee) by using a ruthenium complex and a formic acid/triethylamine mixture under mild conditions.

Summary: The Rh(III)-catalyzed asymmetric reduction of alpha-trifluoromethyl ketones through transfer hydrogenation was developed under mild conditions. It enables a dynamic kinetic resolution process to access a series of enantioenriched cis-trifluoromethyl alcohols. The reaction was performed efficiently in the green solvent 2-MeTHF using HCO2H/Et3N (1 : 1) as the hydrogen source. High yields and excellent diastereo- and enantioselectivities were achieved for the synthesis of CF3-chromanol, CF3-indanol, and CF3-tetralol derivatives.

Summary: In this study, a ruthenium-catalyzed asymmetric transfer hydrogenation of 2,3-disubstituted flavanones was developed. The reaction allowed for the construction of three contiguous stereocenters under basic conditions through a combination of dynamic kinetic resolution and retro-oxa-Michael addition. This method provided chiral flavanols with excellent enantioselectivities and diastereoselectivities.

Catalytic Stereoconvergent Synthesis of Homochiral β-CF3, β-SCF3, and β-OCF3 Benzylic Alcohols

Intriguing multi-chiral molecular architectures can be prepared via ATH for drug discovery.

Summary: A ligand-based approach was used to develop novel PDE5 inhibitors by systematically modifying a trisubstituted pyrazoline scaffold derived from the COX2 inhibitor celecoxib. Novel pyrazolines, such as compound d12, were identified with potent PDE5 inhibitory activity and high selectivity against other PDE isozymes, showing no discernible toxicity and achieving plasma levels >1000-fold higher than IC50 values. These results suggest a novel strategy to inhibit PDE5 with unprecedented potency and isozyme selectivity.

Enantioselectivity in the Noyori-Ikariya Asymmetric Transfer Hydrogenation of Ketones

Summary: This study reveals that there are two spatial regions of the catalyst that simultaneously control the enantioselectivity for any arbitrary substrate: the region of the (tethered) eta(6)-arene ligand and the region of the SO2 moiety. Noncovalent interactions in each region, such as CH-pi, C-H center dot center dot center dot H-C, lone pair-pi, lone pair center dot center dot center dot H-C, play a crucial role in determining the final percent enantiomeric excess (% ee).
