Chemoselective Trimerization of Isocyanides: De Novo Synthesis of 2-Indole-Substituted Quinolines and Pyridines

1-Acryloyl-2-cyanoindole: A Skeleton for Visible-Light-Induced Cascade Annulation

Summary: 1-Acryloyl-2-cyanoindoles were discovered to be novel and efficient scaffolds in visible-light-induced persulfate-promoted cascade cyclization reactions. With this transition-metal-free photocatalytic procedure, various sulfonated/thiocyanated pyrrolo[1,2-a]indolediones were synthesized from 1-acryloyl-2-cyanoindoles with sulfonyl hydrazides/NH4SCN at room temperature under mild reaction conditions.

Rhodium(III)-Catalyzed Synthesis of Diverse Fluorescent Polycyclic Purinium Salts from 6-Arylpurine Nucleosides and Alkynes

Summary: An efficient strategy for assembling a new library of functionalized polycyclic purinium salts with a wide range of anions through Rh-III-catalyzed C-H activation/annulation of 6-arylpurine nucleosides with alkynes under mild reaction conditions is described. The resulting products exhibited tunable photoluminescence across most of the visible spectrum. Mechanistic insights revealed the mode of action of the rhodium catalyst, with a purinoisoquinolinium-coordinated rhodium(I) sandwich complex characterized as the key intermediate.

Synthesis of Fused Pyrimido[1,6-a]indolones via Rhodium(III)-Catalyzed Cascade Annulations

Summary: A novel method for the synthesis of fused pyrimido[1,6-a]-indolone derivatives by annulation of 2-alkynylaryl aldehydes/2-alkynyl-arylidene ketones with N-(pivaloyloxy)-1H-indole-1-carboxamide catalyzed by rhodium has been successfully accomplished. The reaction proceeds through C-H activation based annulation with alkyne moiety followed by addition of nitrogen on to aldehyde/activated alkene to give the products in moderate to good yields. Highly fluorescent dipyrrinone analogues could be synthesized from the derived products.

Rhodium-Catalyzed Atroposelective Construction of Indoles via C-H Bond Activation

Summary: This study reports the rhodium(III)-catalyzed C-H activation of anilines bearing an N-isoquinolyl directing group for oxidative [3+2] annulation with four classes of internal alkynes, leading to atroposelective indole synthesis via dynamic kinetic annulation with C-N reductive elimination constituting the stereo-determining step. The reaction proceeds under mild conditions with high regio- and enantioselectivity and functional group compatibility.

Diarylureas as Antitumor Agents

The first diarylurea to be marketed as an anticancer agent was sorafenib, with others following suit.
