Molecular ontology of the parabrachial nucleus

Samyukta Karthik et al.

Summary: This article presents a developmental-genetic ontology that classifies neurons in the parabrachial nucleus (PB) based on their intrinsic, molecular features, and predicts their neural projections. This molecular framework could accelerate the translation of experimental findings from animal models to human patients.

Netrin1 and reelin signaling are required for the migration of anterolateral system neurons in the embryonic spinal cord

R. Brian Roome et al.

Summary: Projection neurons in the spinal cord dorsal horn transmit pain, itch, and temperature information to the brain through the anterolateral system. Recent research has shown that Phox2a, a transcription factor, is expressed in many developing neurons in this system and has provided insights into their development and wiring of nociceptive circuits. This study reveals that the loss of the axonal guidance and neuronal migration signal netrinl leads to impaired migration of Phox2a+ neurons in the spinal lamina I. Additionally, the absence of Dab1, an intracellular transducer of the neuronal migration signal reelin, impairs the migration of Phox2a+ neurons in the spinal lamina V and lateral spinal nucleus, consistent with deficits in nociception observed in mice with a loss of reelin signaling. Collectively, these findings provide evidence that netrinl and reelin control the development of spinal nociceptive projection neurons and offer a mechanistic explanation for studies linking sequence variations in human genes encoding these neurodevelopmental signals and abnormal pain sensation.

Netrin-1 receptor DCC is required for the contralateral topography of lamina I anterolateral system neurons

Farin B. Bourojeni et al.

Summary: Anterolateral system (AS) neurons relay nociceptive information from the spinal cord to the brain through canonical pathways, with disruption of this pathway leading to altered laterality of spinofugal projections and potential impacts on pain localization precision.

Pain and itch processing by subpopulations of molecularly diverse spinal and trigeminal projection neurons

Racheli Wercberger et al.

Summary: Using retro-TRAP and RNA sequencing, researchers have uncovered extensive molecular diversity of spino-and trigeminoparabrachial projection neurons, including distinct subsets of gene expression. Further investigation revealed significant functional heterogeneity in these projection neurons, showing convergence and segregation of pain-and itch-provoking inputs into molecularly diverse subsets of NK1R- and non-NK1R-expressing neurons.
