Reprogramming of glucocorticoid receptor function by hypoxia

Tineke Vanderhaeghen et al.

Summary: The study found that hypoxia impacts the hepatic response of glucocorticoid receptors to dexamethasone in mice, leading to three types of reprogramming of GR: weaker induction of classical GR-responsive genes, induction of specific genes in hypoxia, and activation of a group of genes by the HPA axis under hypoxia. Additionally, hypoxia-induced activation of the HPA axis is associated with GR-dependent lipolysis and ketogenesis.

Non-genomic steroid signaling through the mineralocorticoid receptor: Involvement of a membrane-associated receptor?

Henk Karst et al.

Summary: The mineralocorticoid receptor (MR) in the mammalian brain mediates both genomic and non-genomic actions, possibly located near or translocated to the cell membrane. Although it is challenging to convincingly visualize membrane localization of endogenous MR or GFP-MR molecules, there is evidence suggesting that MR may be trafficked via beta-arrestin.

Structure of Hsp90-Hsp70-Hop-GR reveals the Hsp90 client-loading mechanism

Ray Yu-Ruei Wang et al.

Summary: Maintaining a healthy proteome is crucial for the survival of all organisms. Hsp90 and Hsp70 are essential molecular chaperones that assist in the folding, remodelling, and maturation of numerous client proteins. The cryo-electron microscopy structure of the GR-loading complex has uncovered the molecular basis of direct coordination by Hsp90 and Hsp70, providing insights into the mechanism of client protein chaperoning.

Cooperativity as quantification and optimization paradigm for nuclear receptor modulators

Pim J. de Vink et al.

Summary: This study proposes a cooperativity mechanism for small molecule modulation of nuclear receptor-protein interactions. By using a thermodynamic model and three-body binding events, the reciprocal effects of nuclear receptor-coregulator binding and nuclear receptor-ligand binding are dissected and quantified. This provides a new conceptual way of thinking about structure-activity relationships for nuclear receptor ligands and has implications for the discovery and optimization of nuclear receptor modulators.

Summary: Different ligands binding to the glucocorticoid receptor (GR) can induce varying conformational changes and biological functions. This study explored the folding pathway of helix 12 (H12) induced by different ligands through molecular dynamics simulations and discovered a novel GR ligand, HP-19, with promising anti-inflammatory activity in the nuclear factor-kappa B signaling pathway and no adverse effects on GR transactivation functions.
