New s-Triazine/Tetrazole conjugates as potent antifungal and antibacterial agents: Design, molecular docking and mechanistic study

Structure-based design, synthesis and antiproliferative action of new quinazoline-4-one/chalcone hybrids as EGFR inhibitors

Summary: A new series of quinazoline-4-one/chalcone hybrids, 7-26, were synthesized and tested as EGFR inhibitors with antiproliferative activity. Three compounds showed the greatest antiproliferative activity and were the most potent EGFR inhibitors. These three compounds improved the levels of active caspase-3, 8, and 9, induced cytochrome c and Bax levels, and down regulated Bcl_2 levels. The most active inhibitors docked well inside EGFR active sites.

Discovery of 2-Phenylquinolines with Broad-Spectrum Anti- coronavirus Activity

Summary: In this study, a compound with potential antiviral activity was identified from a selection of compounds. This compound showed promising activity in the severe phenotypic-based screening assay. Further experiments demonstrated that this compound and its analogues had low toxicity and inhibited the replication of SARS-CoV-2. They also exhibited antiviral activity against other human coronaviruses. Additionally, the compound showed potent inhibition against the SARS-CoV-2 helicase, highlighting its potential against emerging coronavirus outbreaks.

Summary: The development of efficient methods for synthesizing polyfunctional N-heterocycles is important in the fields of organic and medicinal chemistry. Pyrazolo[1,5-a]pyrimidine derivatives have biomedical importance and have been extensively studied for their broad range of biological activities. In recent years, they have also become of interest in materials science due to their photophysical properties. However, there is limited information available on the cyclization of 3(5)-aminopyrazoles and the factors influencing regioselectivity. This study presents a regioselective synthesis of 7-amino-5-arylpyrazolo[1,5-a]pyrimidine-3,6-dicarbonitriles and provides structural confirmation and rationalization for their formation.
