3-Aminopyrazolo[3,4-b]pyridine: Effective Precursor for Barium Hydroxide-Mediated Three Components Synthesis of New Mono- and Bis(pyrimidines) with Potential Cytotoxic Activity

Summary: An efficient one-pot procedure for preparing a new series of pyrazolo[3,4-b]pyridine-fused pyrimidines was developed through the three-component reaction. The synthesized compounds showed promising in vitro cytotoxic activity against MCF-7, HEPG2, and Caco2 cell lines, with hybrid 6h demonstrating the best activity.

Effective synthesis of new benzo-fused macrocyclic and heteromacrocyclic bis(Schiff bases)

Summary: In this study, a new series of macrocyclic bis(Schiff bases), thiamacrocyclic bis(Schiff bases, S-triazole-fused thiamacrocyclic bis(Schiff bases), and [1,2,4]triazolo[3,4-b][1,3,4]thiadiazine-fused macrocycles were synthesized with good to excellent yields. The structures of all new macrocycles were deduced using elemental and spectral data.

Synthesis and antifungal activity of nitrophenyl-pyrazole substituted Schiff bases

Summary: In this study, three new pyrazole-based azomethine isomers were synthesized using environmentally friendly techniques. The addition of a nitrophenyl group on the pyrazole moiety was found to tune the band gap energy and reactivity, leading to increased antifungal activity against C. albicans for the compound bearing the 2-nitrophenyl substituent. Molecular docking simulations also confirmed the strong binding affinity of the most active compound with a model protein.

This study investigated the synthesis of various compounds using pepsin as a biocatalyst. The results showed that by adjusting the reaction conditions, different compounds could be synthesized with good yields, and these compounds exhibited a wide range of antibacterial activity.

Summary: Two series of piperazine-based compounds were synthesized and screened for their acetylcholinesterase inhibitory and DPPH antioxidant activities. The bis(thieno[2,3-b]pyridine) series showed better inhibitory effects, with bis(thieno(2,3-b]pyridine-2-carbonitrile) demonstrating the highest inhibition. Additionally, this compound also displayed good antioxidant activity.

Synthesis, Characterization, and Antimicrobial Evaluation of Novel Thiohydrazonates and Pyrazolo[3,4-b]pyridines

Summary: In this study, 1,3-Diphenyl-1H-pyrazole-4-carbaldehyde and cyanothioacetamide were used to prepare the 2-thioxo-1,2-dihydropyridine-3-carbonitrile derivative, which served as a synthetic precursor. The newly synthesized compounds demonstrated promising in-vitro antimicrobial activities, with thiohydrazonate 7d showing the highest inhibitory activity against all bacterial strains.

New pyrimidine and pyrazole-based compounds as potential EGFR inhibitors: Synthesis, anticancer, antimicrobial evaluation and computational studies

Summary: The study focused on synthesizing new pyrimidines and pyrazoles as tyrosine kinase inhibitors of EGFR. These new compounds showed potent cytotoxicity against human cancer cells, outstanding inhibitory activity against EGFRWT and its mutated isoforms, and broad-spectrum antimicrobial effects. Docking results confirmed their good binding interactions with EGFRWT and EGFRT790M, consistent with the in vitro enzyme assay results. Additional ADMET studies demonstrated their good oral absorption, drug-likeness properties, and low toxicity risks in humans.

Summary: This study aimed to design a new series of selective COX-2 inhibitors with cardioprotective effects, resulting in the discovery of compound 7a with high selectivity and anti-inflammatory activity. In vitro evaluation against COX-1 and COX-2 enzymes confirmed the cardiovascular benefits of compound 7a over celecoxib. Docking simulation and ADME study supported the potential use of compound 7a as a new oral anti-inflammatory agent.

Efficient synthesis and characterization of novel bis(chromenes) and bis(benzo[f]chromenes) linked to thiazole units

Summary: New carbothioamides, linked to chromene or benzo[f]chromene units, were synthesized and used to prepare various substituted thiazoles and thiazol-4(5H)-ones through reactions with different alpha-halocarbonyl compounds and bis(alpha-bromoketones). Moreover, condensation of thiazol-4(5H)-ones with appropriate bis(aldehydes) yielded bis(thiazol-4(5H)-ones) linked to chromene or benzo[f]chromene units via different cores.

Novel nicotinonitrile-coumarin hybrids as potential acetylcholinesterase inhibitors: design, synthesis, in vitro and in silico studies

Summary: In this study, a new series of nicotinonitrile-coumarin hybrids were designed as potential acetylcholinesterase inhibitors, showing more effective inhibitory activity than the standard drug donepezil. These hybrids also exhibited good free radical-scavenging capabilities and demonstrated anti-cytotoxic activity in vitro. Additionally, docking studies revealed excellent interactions between the nicotinonitrile-coumarin hybrids and AChE.
