Development of Sustained Release Baricitinib Loaded Lipid-Polymer Hybrid Nanoparticles with Improved Oral Bioavailability

Summary: This study prepared lipid-polymer hybrid nanoparticles loaded with baricitinib and evaluated their physicochemical properties, morphology, in vitro release, and in vivo pharmacokinetics. The optimized formulation, B-PLN4, showed promising results with high entrapment efficiency and sustained release pattern. It significantly enhanced the bioavailability of baricitinib compared to the pure suspension. These findings suggest that lipid-polymer hybrid nanoparticles could be a promising drug delivery option for improving the pharmacokinetics of baricitinib.

Efficacy and safety of baricitinib plus standard of care for the treatment of critically ill hospitalised adults with COVID-19 on invasive mechanical ventilation or extracorporeal membrane oxygenation: an exploratory, randomised, placebo-controlled trial

Summary: This study evaluated the efficacy and safety of baricitinib plus standard of care in critically ill COVID-19 patients. The results showed that compared to placebo, treatment with baricitinib significantly reduced mortality at both 28 and 60 days. Additionally, there were no significant differences in ventilator-free days and hospitalization duration between the baricitinib and placebo groups.

The results show that these complexes have the potential to be used as thermo-sensitive nanocarriers with controlled release properties.

Effect of Baricitinib on TPA-induced psoriasis like skin inflammation

Summary: In this study, topical application of Baricitinib showed promising therapeutic effects on chronic TPA-induced psoriasis in mice, effectively inhibiting inflammation and promoting skin repair. Baricitinib significantly reduced ear swelling, leukocyte infiltration, cell proliferation, and angiogenesis, while also decreasing the phosphorylation of STAT3 and STAT1. These results suggest that Baricitinib could be a potential topical treatment for the progression of psoriasis.
