Mitochondrial copper depletion suppresses triple-negative breast cancer in mice

Liyang Cui et al.

Summary: The research developed a mitochondria-targeted copper-depleting nanoparticle that effectively inhibited the growth of triple-negative breast cancer cells and improved survival rates in mouse models. By depriving copper within cancer cells, this approach induced metabolic changes and apoptosis, highlighting its potential clinical relevance.

Triphenylphosphonium-conjugated glycol chitosan microspheres for mitochondria-targeted drug delivery

Young Hwa Lee et al.

Summary: A vector for mitochondria-targeted drug delivery was developed by synthesizing TPP-modified glycol chitosan polymeric microspheres. The microspheres possess the potential to effectively deliver drugs to mitochondria. Various analysis and experiments confirmed their efficacy as drug delivery carriers.

Multifunctional Organic Fluorescent Probe with Aggregation-Induced Emission Characteristics: Ultrafast Tumor Monitoring, Two-Photon Imaging, and Image-Guide Photodynamic Therapy

Haijun Ma et al.

Summary: The development of multifunctional photosensitizers with aggregation-induced emission properties is essential for advancing photodynamic therapy. In this study, a multifunctional PS named DSABBT NPs was designed and prepared to provide ultrafast staining, excellent two-photon bioimaging, and high-efficiency image-guided PDT. DSABBT NPs exhibited bright solid-state fluorescence, high two-photon absorption cross-section, fast staining and imaging capabilities, as well as efficient singlet oxygen generation and image-guided PDT efficiency. Overall, DSABBT NPs show promise as multifunctional photosensitizers for cancer treatment.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Nuclear sensing of breaks in mitochondrial DNA enhances immune surveillance

Marco Tigano et al.

Summary: The study reveals that mtDSBs activate a type-I interferon response, leading to the phosphorylation of STAT1 and activation of interferon-stimulated genes. After breaks in mtDNA, herniation mediated by BAX and BAK releases mitochondrial RNA into the cytoplasm and triggers a RIG-I-MAVS-dependent immune response.

Parallel Genomic Alterations of Antigen and Payload Targets Mediate Polyclonal Acquired Clinical Resistance to Sacituzumab Govitecan in Triple-Negative Breast Cancer

James T. Coates et al.

Summary: This study identified mechanisms of resistance to the antibody-drug conjugate (ADC) SG in different metastatic subclones of an individual patient, revealing genomic alterations in both antibody and drug payload targets. Experiments also demonstrated that a TROP2 gene mutation can confer resistance to SG.

An ROS-sensitive tegafur-PpIX-heterodimer-loaded in situ injectable thermosensitive hydrogel for photodynamic therapy combined with chemotherapy to enhance the tegafur-based treatment of breast cancer

Zhiqiang Zhang et al.

Summary: Researchers prepared a biomimetic and thermosensitive hydrogel encapsulating reactive oxygen species (ROS)-sensitive tegafur (TF)-protoporphyrin IX (PpIX) heterodimers (TTP) to synergize chemotherapy and photodynamic therapy for breast cancer treatment. This innovative approach overcame major obstacles in breast cancer treatment and opened up new paths for treatment.
