Lysosomal lipoprotein processing in endothelial cells stimulates adipose tissue thermogenic adaptation

Alexander W. Fischer et al.

Summary: This study reveals that vascular endothelial cells in adipose tissues internalize significant amounts of triglyceride-rich lipoprotein particles under cold exposure which lead to enhanced proliferation of endothelial cells and adipocyte precursors, ultimately impacting thermogenic capacity through a β-oxidation-dependent mechanism.

Case Report: PNPLA2 Gene Complex Heterozygous Mutation Leading to Neutral Lipid Storage Disease With Myopathy

Jiejing Shi et al.

Summary: Neutral lipid storage disease with myopathy (NLSDM) caused by PNPLA2 gene mutation is a rare clinical myopathy with characteristic changes in skeletal muscle imaging and pathology. The main symptoms include muscle weakness and muscular atrophy. Detection of PNPLA2 gene mutation is crucial for diagnosis, with asymmetry and progressive limb weakness being key clinical features. Muscle MRI primarily shows involvement in the posterior group of the lower limbs. Jordans bodies in the peripheral blood smear and coarse-grained lipid deposits with rimmed vacuoles in muscle fibers are characteristic pathological findings.

Sphingolipids in metabolic disease: The good, the bad, and the unknown

Christopher D. Green et al.

Summary: The dysregulation of sphingolipid metabolism is associated with the pathogenesis of metabolic diseases, with ceramide exacerbating metabolic diseases by regulating signaling and metabolic pathways, while its metabolite S1P and its receptors have multifaceted and sometimes opposing actions in tissues.

Eruptive xanthoma model reveals endothelial cells internalize and metabolize chylomicrons, leading to extravascular triglyceride accumulation

Ainara G. Cabodevilla et al.

Summary: Despite the primary role of LpL in tissue uptake of fatty acids from chylomicrons, studies have shown that an additional pathway exists for transferring dietary lipids to tissues. This pathway involves endothelial cell uptake of chylomicrons, hydrolysis in lysosomes, and subsequent lipid accumulation in cells. Competing with HDL and targeting the scavenger receptor-BI can inhibit this process in vivo, potentially reducing lipid accumulation in tissues and preventing conditions such as eruptive xanthomas.
