Complementarity of neutron, XFEL and synchrotron crystallography for defining the structures of metalloenzymes at room temperature

Tadeo Moreno-Chicano et al.

Summary: Room-temperature macromolecular crystallography allows protein structures to be determined under close-to-physiological conditions, but can present challenges, especially for metalloenzymes. This study describes and compares the structures of a multifunctional globin, dehaloperoxidase B (DHP-B), obtained using different methods of room-temperature crystallographic structure determination. The results show the differences in sample requirements, data-collection time, and potential for radiation damage between these methods.

Summary: Dehaloperoxidase (DHP) is a multi-functional catalytic globin found in a marine worm. Its physiological functions include oxygen transport and oxidation of toxic substances. The electronic structure of DHP varies in different states, with pH playing a role in modulating its electron-donating properties.

Summary: The hemoglobin from the marine worm Amphitrite ornata has multiple enzymatic activities in addition to its O-2-transport function, where the globin, named dehaloperoxidase (DHP), serves as a multifunctional catalytic hemoglobin. DHP employs four mechanisms for substrate oxidation as a defense mechanism against toxic metabolites and is particularly emphasized for its peroxygenase activity.
