Summary: The tissue repairing capacity and immunomodulatory effects of mesenchymal stem cells (MSCs) have been utilized for treating various inflammatory disorders, but low survival rates after transplantation limit their clinical potential; a novel strategy involving heterospheroids (HS) comprising MSCs and quercetin-loaded microspheres (MSCHS) was designed to improve the microenvironment of MSCs and achieve better therapeutic outcomes in a murine model of colitis. MSCHS displayed enhanced survival capacity and better therapeutic outcomes compared to 3D-cultured MSCs (MSC3D), partially due to their superior PGE2 secretion in promoting tissue repair.
