Structural optimizations and bioevaluation of N-substituted acridone derivatives as strong topoisomerase II inhibitors

Targeting specific genes and proteins involved in the growth and survival of cancer cells has become a top priority in global research. Indole moiety, a combination of aromatic-heterocyclic compounds, has emerged as a promising scaffold for the development of anticancer drugs due to its bioavailability, unique chemical properties, and significant pharmacological behaviors. Recent advances in the medicinal chemistry of indole derivatives, including the synthesis of potential anticancer compounds and their mechanism of action, are discussed in this review.

Design and discovery of new 1,2,4-triazolo[4,3-c]quinazolines as potential DNA intercalators and topoisomerase II inhibitors

Summary: A new series of compounds were designed and synthesized as Topo II inhibitors and DNA intercalators, showing high cytotoxic activities against cancer cell lines. The most promising candidates were evaluated for their Topo II inhibition and DNA intercalation abilities, indicating a potential mechanism for their antiproliferative effects. The most promising compound was found to induce apoptosis and cell cycle arrest in cancer cells, while molecular docking studies suggested that DNA binding and/or Topo II inhibition may contribute to the cytotoxicity of the synthesized compounds.

Design, synthesis, molecular docking and in silico ADMET profile of pyrano [2,3-d]pyrimidine derivatives as antimicrobial and anticancer agents

Summary: Pyrano[2,3-d]pyrimidine derivatives showed promising antimicrobial and cytotoxic activities against various microbial strains and cell lines, exhibiting strong inhibitory effects on cell wall synthesis and DNA intercalation. The compounds also demonstrated higher anticancer activities than doxorubicin, with some derivatives showing potent Topoisomerase II inhibitory activities. Molecular modeling studies supported the in vitro results, indicating the potential of these derivatives for further development as anti-infective and anticancer agents.

Discovery of novel triazolophthalazine derivatives as DNA intercalators and topoisomerase II inhibitors

Summary: The designed and synthesized triazolophthalazine derivatives showed promising cytotoxic activities against human cancer cell lines by acting as Topo II inhibitors and DNA intercalators, with compound IXb being the most potent counterpart. Additionally, compound IXb demonstrated excellent DNA binding affinity and significant Topo II inhibitory activity, inducing apoptosis and affecting the cell cycle. Molecular docking studies revealed the binding patterns of the designed compounds against the DNA-Topo II complex and DNA.

Experimental and computational study of naphthalimide derivatives: Synthesis, optical, nonlinear optical and antiviral properties

Summary: Novel 5-nitro-2-(substitutedphenyl)benzoxazole and 2-(substitutedphenyl)oxazolo[4,5-b]pyridine derivatives were designed and synthesized as potential anticancer agents, with two compounds showing promising inhibitory activity against hTopo II alpha. However, these compounds did not exhibit satisfactory cytotoxic activity on various cancer cell lines. Molecular docking and ADME/Tox studies were conducted to understand the mechanism of inhibition and predict drug-likeness and pharmacokinetic properties, respectively.
