This study found that CD36 expression in tumor-infiltrating CD8(+) T cells, induced by TME cholesterol, is associated with tumor progression and poor survival, and that genetic ablation of Cd36 in these T cells leads to enhanced tumor eradication. Targeting CD36 or inhibiting ferroptosis could restore T cell function and enhance antitumor efficacy, especially in combination with anti-PD-1 antibodies.

Human CD36hi monocytes induce Foxp3+ CD25+ T cells with regulatory functions from CD4 and CD8 subsets

Summary: The study reveals that a subset of CD14(+) monocytes can generate regulatory Foxp3(+) T-bet(+) T cells from umbilical cord blood, which suppress T-cell proliferation and ameliorate graft-versus-host disease. Additionally, adult peripheral blood monocytes are capable of inducing Foxp3(+) T cells, but their induction is inhibited by lymphoid cells from adult peripheral blood in neonates. This suggests a novel immunoregulatory role of monocytes in generating regulatory T cells with implications for both neonates and adults.

Inhibiting lipid synthesis dependent on SREBPs in T-reg cells can enhance antitumor immune responses without autoimmune toxicity, and deletion of SCAP in these cells inhibits tumor growth and enhances immunotherapy. SCAP and SREBPs signaling regulate lipid synthesis and inhibitory receptor signaling in T-reg cells, pointing to new ways of targeting these cells for cancer therapy.
