Epithelial-Mesenchymal Transition (EMT) as a Therapeutic Target

Sven Jonckheere et al.

Summary: Metastasis, the spread of cancer cells to distant sites, is the primary cause of cancer morbidity and mortality. During the initial stages of metastasis, cancer cells undergo EMT, becoming migratory and invasive mesenchymal-like cells with stem cell properties. Targeting EMT in therapy has the potential to prevent cancer cell invasion and dissemination, reduce cancer stemness, and enhance the effectiveness of chemotherapy.

Could inhibition of metalloproteinases be used to block the process of metastasis?

Jesus Alba et al.

Summary: Metastasis is a multisequential process that involves the migration of tumor cells to distant tissues from the primary tumor. This process requires the transformation of tumor epithelial cells into mesenchymal cells (EMT) and the degradation of the extracellular matrix (ECM). This review focuses on the different options to inhibit MMPs, with a specific emphasis on the role of MMP7 in the metastatic processes of various tumors.

The long non-coding RNA ET-20 mediates EMT by impairing desmosomes in breast cancer cells

Meera Saxena et al.

Summary: The study identified 114 novel lncRNAs associated with EMT, with ET-20 being regulated by the EMT master transcription factor Sox4 and playing a role in promoting EMT by interacting with desmosomal proteins. ET-20 was found to be upregulated in invasive breast cancer cell lines and targeting it could be a potential therapeutic approach to inhibit EMT and breast cancer metastasis.

Cancer drug resistance induced by EMT: novel therapeutic strategies

Javier De las Rivas et al.

Summary: This review explores the important clinical benefits achieved in cancer patients through drug-targeting strategies, but drug resistance remains a major issue. The impact of epithelial-mesenchymal plasticity and tumor microenvironment on treatment effectiveness is discussed, along with the use of bioinformatics and pharmacogenomics to study the biological effects of epithelial-to-mesenchymal transition on drug resistance and develop novel pharmacological approaches in the future.

miR-30a/SOX4 Double Negative Feedback Loop is modulated by Disulfiram and regulates EMT and Stem Cell-like properties in Breast Cancer

Zijian Liu et al.

Summary: miR-30a inhibits EMT and CSC phenotypes in breast cancer cells by targeting SOX4 and forming a double-negative feedback loop. Knockdown of SOX4 suppresses EMT and CSC phenotypes through the TGF-beta/SMAD pathway, consistent with the effects of miR-30a overexpression. High expression of miR-30a is associated with a good prognosis in breast cancer patients.

Linking EMT programmes to normal and neoplastic epithelial stem cells

Arthur W. Lambert et al.

Summary: Epithelial stem cells play critical roles in tissue generation, maintenance, and repair, while cancer stem cells are implicated in the growth and recurrence of many carcinomas. The activation of epithelial-mesenchymal transition (EMT) is linked to the formation of both normal and neoplastic stem cells, but the mechanistic basis of this connection remains unclear. Understanding the elusive links between EMT programs and the stem cell state will advance our knowledge of normal stem cell biology and cancer pathogenesis.
