Neoadjuvant Chemotherapy and Immunotherapy in Luminal B-like Breast Cancer: Results of the Phase II GIADA Trial

Maria Vittoria Dieci et al.

Summary: This study explored the role of immunotherapy in hormone receptor-positive, HER2-negative breast cancer. The results showed that Luminal B-like breast cancer patients had a higher rate of pathologic complete response to sequential anthracyclines and anti-PD-1 treatment. Tumor-infiltrating lymphocytes, immune-related gene-expression signatures, and specific immune cell subpopulations were closely associated with pathologic complete response.

Impact of HPV status on immune responses in head and neck squamous cell carcinoma

Hannan A. Qureshi et al.

Summary: The impact of immune cell composition and tumor reactivity on HPV-positive and HPV-negative head and neck squamous cell carcinoma was studied. Results showed significant differences in immune features based on HPV status, and increased B cell and plasma cell content in tumors were associated with improved progression-free survival in HPV-negative patients treated with immune checkpoint inhibitors.

Tumor-specific T cells support chemokine-driven spatial organization of intratumoral immune microaggregates needed for long survival

Ziena Abdulrahman et al.

Summary: This study investigates the tumor immune microenvironment associated with tumor-specific T-cell responses and finds that IR+ patients have higher survival rates and immune cell infiltration, while IR- patients exhibit the presence of immunosuppressive cells. Tumor-specific T cells in IR+ tumors produce chemokines to sustain immune cell aggregation, which is associated with good survival after standard therapy.

Sex and cancer immunotherapy: Current understanding and challenges

Laura Pala et al.

Summary: Recent evidence suggests that patients' sex plays a significant role in the antitumor immune response, with complex interactions among hormones, genes, behaviors, and the microbiome affecting innate and adaptive immune functions as well as immune evasion mechanisms. These interactions ultimately impact the efficacy and toxicity of immune checkpoint inhibitors in solid tumors.

Summary: In head-and-neck squamous cell carcinoma (HNSCC), the expression patterns of sex hormone receptors differ between HPV-positive and HPV-negative patients. Estrogen receptor alpha is associated with HPV-positive tumors, while androgen and progesterone receptors are correlated with poor patient prognosis.

Characterization of the tumor immune microenvironment in human papillomavirus-positive and -negative head and neck squamous cell carcinomas

Farah Succaria et al.

Summary: Approximately 15% of advanced head and neck squamous cell carcinomas respond to anti-PD-(L)1 monotherapies, with tumor PD-L1 expression and human papillomavirus (HPV) status proposed as biomarkers to identify patients likely to benefit. The study found that HPV-positive HNSCC tumors had significantly higher densities of certain immune cells compared to HPV-negative tumors, and that there was no variation in PD-L1 expression patterns based on tumor viral status.
