Antitumor activity and molecular mechanism of proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibition

Huimin Sun et al.

Summary: Proprotein convertase subtilisin/kexin type 9 (PCSK9) plays an important role in regulating cholesterol metabolism and lipoprotein homeostasis. In addition to its role in cardiovascular diseases, PCSK9 has been found to be involved in tumor development and progression. Inhibition of PCSK9 can attenuate tumor progression and promote apoptosis of cancer cells.

Summary: This study investigates the immune response to hepatitis B vaccination and finds similar kinetics between circulating Tfh cells and splenic Tfh cells; cTfh cells are positively correlated with anti-HBs at one week after vaccination; in vitro experiments show preferential activation of CXCR3(-)Tfh cell subsets in peripheral blood mononuclear cells stimulated by HBsAg.

EMT, cancer stem cells and autophagy; The three main axes of metastasis

Ghader Babaei et al.

Summary: Epithelial-mesenchymal transition (EMT) and Cancer stem-like cells (CSCs) are major factors contributing to cancer cell metastasis. Autophagy has recently gained attention, playing a dual role in cancer cell development depending on cancer type, tissue, and stage. Studies show different signaling pathways are involved in cancer stem cells, autophagy, and EMT. Investigating effective signaling pathways in all three processes could help determine appropriate therapeutic goals.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

PCSK9 and cancer: Rethinking the link

Khadijeh Mahboobnia et al.

Summary: PCSK9 plays an important role in the development and progression of various cancers, highlighting its potential significance in cancer treatment. Inhibiting PCSK9 may help suppress the proliferation and invasion of tumor cells. However, the current lack of data on the role of PCSK9 in cancer and the impact of PCSK9 inhibition in cancer therapy limits our understanding of its effectiveness and safety.

Pro-protein convertase subtilisin/kexin type 9 promotes intestinal tumor development by activating Janus kinase 2/signal transducer and activator of transcription 3/SOCS3 signaling in ApcMin/+ mice

Kai Yang et al.

Summary: The study demonstrated that PCSK9 may have a potential oncogenic role in the development and progression of colorectal cancer in vivo via activation of JAK2/STAT3/SOCS3 signaling pathway. Overexpression of PCSK9 led to increased adenomas and adenocarcinomas in mice, while treatment with evolocumab reduced tumor growth and prevented the development of adenocarcinomas.
