Betulinic acid-nucleoside hybrid prevents acute alcohol -induced liver damage by promoting anti-oxidative stress and autophagy

Summary: Alcoholic abuse is a major cause of liver diseases globally, with oxidative stress being a critical factor. This study showed that BAN, a betulinic acid and nucleoside hybrid compound with good water solubility, effectively reversed acute alcohol-induced liver damages by reducing serum ALT level and liver oxidative stress. The protective role of BAN was mediated through hepatic FXR activation and the promotion of cellular autophagy.

Effect of betulinic acid administration on TLR-9/NF-κB /IL-18 levels in experimental liver injury

Summary: This study investigated the potential protective effects of betulinic acid on APAP-induced hepatotoxicity in rats. The results showed that betulinic acid significantly decreased the levels of TLR9, IL-18, NF-kappa B, and MDA in liver injury groups. This suggests that betulinic acid may be an effective agent for the prevention and treatment of acute liver diseases.

Summary: In this study, it was found that EZH2 and JMJD3 antagonistically modulate HSCs activation. The therapeutic effects of DZNep as an epigenetic drug in liver fibrosis are associated with the regulation of EZH2 towards direct target genes encoding TGF-I31 pseudoreceptor BAMBI, anti-inflammatory cytokine IL10 and cell cycle regulators CDKN1A, GADD45A and GADD45B, which are also regulated by JMJD3. This study provides new mechanistic insight into the epigenetic modulation of EZH2 and JMJD3 in HSCs biology and hepatic fibrogenesis.
