Engineered ionizable lipid siRNA conjugates enhance endosomal escape but induce toxicity in vivo

Sixteen Different Types of Lipid-Conjugated siRNAs Containing Saturated and Unsaturated Fatty Acids and Exhibiting Enhanced RNAi Potency

Summary: Lipid-conjugated siRNAs showed excellent cellular uptake efficiency and superior RNAi potency, with the conjugates composed of 16-18 carbon chains as fatty acids demonstrating particularly strong effects. Additionally, conjugate siRNAs containing 18 carbon chains with a cis-form were found to have better RNAi potency compared to those with a trans-form. These lipid-siRNAs have the potential to address the challenges hindering the clinical application of siRNAs.
