Coactivator-associated arginine methyltransferase 1 controls oligodendrocyte differentiation in the corpus callosum during early brain development

Yugo Ishino et al.

Summary: Protein arginine methylation, mediated by Carm1, plays a crucial role in regulating cellular events such as cell proliferation and differentiation. In this study, Carm1 was found to be highly expressed in oligodendrocyte lineage cells in the developing brain, and its inhibition resulted in reduced proliferation and impaired differentiation of these cells. Carm1 activity was also found to be necessary for the appropriate formation of myelin processes and myelin sheaths, suggesting its essential role in the development of oligodendrocytes.

Hypoxic stress increases NF-κB and iNOS mRNA expression in normal, but not in keratoconus corneal fibroblasts

Tanja Stachon et al.

Summary: The study investigates the effect of hypoxia through NF-kappa B signaling pathways on iNOS, HIF, ROS, and proliferation of normal and KC human corneal fibroblasts in vitro. Hypoxia increases NF-kappa B and iNOS mRNA expression in normal HCFs, but not in KC-HCFs, possibly due to preexisting oxidative stress. HIF and PHD2 do not show altered iNOS regulation under hypoxic conditions in KC-HCFs, suggesting they may not play a role in keratoconus pathogenesis.

PRMT4 inhibitor TP-064 inhibits the pro-inflammatory macrophage lipopolysaccharide response in vitro and ex vivo and induces peritonitis-associated neutrophilia in vivo

Yiheng Zhang et al.

Summary: The PRMT4 inhibitor TP-064 induces peritonitis-associated neutrophilia in vivo and inhibits the pro-inflammatory macrophage lipopolysaccharide response in vitro and ex vivo. These findings suggest that TP-064 may be a potential therapy for NF-kappa B-based inflammatory diseases.

PRMT1 enhances oncogenic arginine methylation of NONO in colorectal cancer

Xin-Ke Yin et al.

Summary: Arginine methylation, catalyzed by PRMTs, plays a crucial role in CRC. Overexpression of NONO in CRC tissue is associated with poor prognosis. Methylation of NONO by PRMT1 at R251 promotes CRC growth and metastasis, indicating PRMT1 inhibition as a potential therapeutic strategy for CRC treatment.

Loss of cholinergic innervation differentially affects eNOS-mediated blood flow, drainage of Aβ and cerebral amyloid angiopathy in the cortex and hippocampus of adult mice

Shereen Nizari et al.

Summary: The interplay between cholinergic innervation and vascular function is crucial in the development and progression of CAA, suggesting that combined eNOS/cholinergic therapies may enhance Aβ clearance from the brain and reduce its deposition as CAA.

Risk Factors for Alzheimer's Disease: An Epidemiological Study

Jana Janoutova et al.

Summary: The study conducted two epidemiological projects to investigate potential genetic, vascular, and psychosocial risk factors for Alzheimer's disease. Results showed a higher prevalence of AD in females and individuals with lower education levels, as well as a higher proportion of manual workers among cases. Coronary heart disease was found to be more common in AD cases compared to controls. Early detection and understanding of the risk factors for AD may be beneficial for prevention and treatment of the condition.

Decreased Expression and Uncoupling of Endothelial Nitric Oxide Synthase in the Cerebral Cortex of Rats with Thioacetamide-Induced Acute Liver Failure

Krzysztof Milewski et al.

Summary: Acute liver failure leads to deregulated nitric oxide signaling in the brain, resulting in reduced NOS activity and cofactor BH4 content, as well as eNOS uncoupling. These changes are associated with increased reactive oxidative species and decreased cerebral cortical blood flow, indicating a potential causal relationship between eNOS dysfunction and neurologic complications of ALF.

Histone Arginine Methyltransferase CARM1-Mediated H3R26me2 Is Essential for Morula-to-Blastocyst Transition in Pigs

Zubing Cao et al.

Summary: The study revealed that CARM1 specifically catalyzes H3R26me2 in porcine embryos to promote blastocyst formation and participates in blastocyst development. Functional studies using chemical inhibition and RNA interference demonstrated that inhibition or knockdown of CARM1 severely compromised blastocyst development and altered the transcriptome characterized by downregulation of key genes associated with Hippo and PI3K-AKT signaling pathways.
