Summary: Systemic sclerosis is a complex disease characterized by fibrosis in the skin and multiple internal organs. Understanding its pathogenesis and progression is a pressing scientific challenge. Recent studies have identified toll-like receptor 4 (TLR4) signaling as a key driver of fibrosis in systemic sclerosis, with TLR4 sensing danger signals and promoting fibrogenesis. Tenascin-C, an extracellular matrix glycoprotein, is highly upregulated in systemic sclerosis and plays a role in driving fibroblast activation and fibrosis progression.

Summary: TGF-beta isoforms play a crucial role in regulating tissue homeostasis and immune cell function, while Tenascin family members are able to modulate the bioavailability of TGF-beta through their C-terminal domains. This regulation likely involves a conformational change leading to activation of the latent cytokine and subsequent effects on cell responses, such as cytostasis and epithelial-to-mesenchymal transition in mammary epithelial cells. These findings suggest a novel common function for the Tenascin family in maintaining tissue homeostasis under both healthy and pathological conditions.

Tenascin-C in Heart Diseases-The Role of Inflammation

Summary: Tenascin-C is a large ECM glycoprotein transiently expressed in the heart during embryonic development, but rarely detected in normal adults, with its expression strongly up-regulated in inflammation. TNC appears to worsen adverse ventricular remodeling by aggravating inflammation/fibrosis, and high levels predict poor prognosis in patients with various heart diseases, making it a potential biomarker for disease stratification and therapy selection.
