Phosphatidylinositol phosphates modulate interactions between the StarD4 sterol trafficking protein and lipid membranes

Summary: This study demonstrates a PIP-selective mechanism by which the protein StarD4 extracts cholesterol from liposomes. The lipid composition of the donor and acceptor membranes plays a role in modulating StarD4-mediated transport. Molecular dynamics simulations reveal the key interaction sites of StarD4 with PIP-containing membranes. These findings provide insights into the mechanism by which PIPs regulate cholesterol transfer activity via StarD4.

A Unique Junctional Interface at Contact Sites Between the Endoplasmic Reticulum and Lipid Droplets

Summary: Lipid droplets (LDs) store metabolic energy in the form of neutral lipids and originate from the endoplasmic reticulum (ER), maintaining contact with it to facilitate protein and lipid exchange. Proper formation of ER-LD junctions is crucial to prevent the formation of aberrant LDs, which can impact lipid homeostasis and lead to lipid storage diseases. The close contacts between LDs, peroxisomes, and mitochondria play a role in metabolic channeling of fatty acids, highlighting the importance of LD contact sites in lipid metabolism.

Highly accurate protein structure prediction for the human proteome

Summary: Using the AlphaFold method, the structural coverage of the proteome has been significantly expanded, covering 98.5% of human proteins with 58% of residues having confident predictions and 36% having very high confidence. Introducing new metrics to interpret the dataset and identify disordered regions, this study aims to provide high-quality predictions for generating biological hypotheses.
