Characterization of an angular domain fluorescence optical projection tomography system for mesoscopic lymph node imaging

Summary: Transmittance and fluorescence optical projection tomography can provide high-resolution and high-contrast visualization of whole biological specimens, but their applications are limited by light scattering in samples. A new angle-restricted transmittance/fluorescence system was presented and characterized in terms of reconstruction accuracy, spatial resolution, depth of focus, and fluorescence limits of detection. Results using lymph node mimicking phantoms showed promising capabilities for clinical lymph node applications.

Physical and chemical mechanisms of tissue optical clearing

Summary: Advanced optical methods combined with various probes have enabled molecular imaging within living cells, but improving imaging resolution to the subcellular level is a major challenge for larger tissues and in vivo studies. Tissue optical clearing techniques offer innovative ways to perform deep-tissue imaging, with various methods developed based on similar physical principles but using different chemical approaches. This includes refractive index matching and chemical treatments to reduce scattering and absorption for deeper tissue imaging.
