Drug Resistance Mechanism of M46I-Mutation-Induced Saquinavir Resistance in HIV-1 Protease Using Molecular Dynamics Simulation and Binding Energy Calculation

Nilottam Rana et al.

Summary: The study investigates the molecular mechanism of saquinavir resistance caused by the M46I mutation in HIV-1 protease. The results show that this mutation significantly affects the protein's structure and stability, leading to decreased van der Waals interaction and binding free energy, as well as inward flap curling and wider opening. These findings provide valuable insights for the development of inhibitors against drug-resistant HIV-1 protease.

Summary: The development of GSIs and GSMs for Alzheimer's disease and cancer treatment presents a promising therapeutic opportunity. Cryo-electron microscopy structures reveal that these inhibitors and modulators target specific regions of gamma-secretase, interfering with substrate recruitment. Shared themes and variations in inhibitor and modulator recognition may guide the development of next-generation substrate-selective inhibitors.

Summary: The study investigated the inhibitory effects of phytochemicals from Indian medicinal plants on SARS-CoV-2 drugable targets through molecular docking, molecular dynamics simulation, and MM-PBSA analysis. Isoquercetin and 10-Hydroxyaloin A were found to have significant efficacy in binding to the active site of SARS-CoV-2 Mpro. Further experimental validation is needed to assess their anti-SARS-CoV-2 potential.

Kumari Sunita Prajapati et al.

Summary: This study identified 22 common miRNAs involved in regulating major stem cell signaling pathways, including miRNAs associated with breast cancer stemness. The clinical relevance of these miRNAs was confirmed by survival analysis in breast cancer patients, showing poor overall survival and high expression levels in invasive breast cancer subgroups. Further validation in in vitro models demonstrated the downregulation of lead miRNA with phytochemical treatment.
