Methyl beta-D-galactopyranoside esters as potential inhibitors for SARS-CoV-2 protease enzyme: synthesis, antimicrobial, PASS, molecular docking, molecular dynamics simulations and quantum computations

Summary: Carbohydrate esters play an important role in medicinal chemistry for the synthesis of biologically active drugs. In this study, methyl beta-D-galactopyranoside (MGP) was reacted with various acyl halides to produce 6-O-acyl MGP esters, which were further modified into 2,3,4-tri-O-acyl MGP esters. The newly synthesized compounds showed promising antifungal functionality compared to their antibacterial activities. Among them, compounds 3 and 10 exhibited the strongest inhibitory effects against Bacillus subtilis and Escherichia coli. Density functional theory calculations and molecular docking revealed the properties and interactions of these compounds. Molecular dynamics simulation validated the binding affinities obtained from molecular docking. The pharmacokinetic characterization demonstrated that these MGP esters have improved kinetic properties. Structure activity relationships study indicated that the acyl chain and chemical groups influenced the activity of the esters. Overall, this comprehensive computational and statistical analysis suggests that these selected MGP esters could be potential inhibitors against SARS-CoV-2.

Synthesis and biological evaluation of novel aryloxyacetic acid hydrazide derivatives as anticancer agents
