Polyamines in cancer: integrating organismal metabolism and antitumour immunity

Cassandra E. Holbert et al.

Summary: Polyamines play a crucial role in the initiation and progression of cancer, and dysregulation of their metabolism is common in many cancers. Recent research suggests that polyamines can affect the antitumour immune response and contribute to the lack of response to immune checkpoint blockade in immunologically 'cold' tumours. Furthermore, the interaction between the microbiota and tissues can significantly alter polyamine metabolism, content, and function.

Enriched metabolites that potentially promote age-associated diseases in subjects with an elderly-type gut microbiota

Shin Yoshimoto et al.

Summary: The study compared fecal metabolites between age-matched and age-mismatched elderly subjects and found significant differences in metabolites associated with age-related diseases. Metabolites enriched in the age-matched elderly group were more likely to be risk factors for age-associated diseases such as arteriosclerosis and colorectal cancer.

Harnessing the polyamine transport system to treat BRAF inhibitor-resistant melanoma

Eric T. Alexander et al.

Summary: BRAF inhibitors are effective in treating melanoma but recurrences are common. A novel compound, AP, has shown promise in delaying recurrence and reducing tumor-promoting macrophages in BRAF inhibitor-resistant melanoma. AP targets polyamine transport system in BRAF inhibitor-resistant CSCs and blocks CXCR4 signaling in invasive melanoma cells and pro-tumorigenic macrophages.

Histone deacetylase 10, a potential epigenetic target for therapy

Fajuan Cheng et al.

Summary: HDAC10, a class II family member, has been implicated in various tumors and non-tumor diseases, playing crucial roles in regulating cellular processes. Studying the biological functions of HDAC10 is essential for the future development of specific inhibitors.

Symbiotic polyamine metabolism regulates epithelial proliferation and macrophage differentiation in the colon

Relationships of Gut Microbiota Composition, Short-Chain Fatty Acids and Polyamines with the Pathological Response to Neoadjuvant Radiochemotherapy in Colorectal Cancer Patients

Lidia Sanchez-Alcoholado et al.

Summary: This study suggests that gut microbiota composition plays a role in the response to cancer therapies in CRC patients. Responders had higher microbial diversity and abundance of butyrate-producing bacteria, as well as higher levels of specific metabolites, while non-responders exhibited a microbiota rich in pro-inflammatory species.Ultimately, a baseline consortium of five bacterial species was identified as potential predictors of CRC treatment outcome.
