Structural basis of TRPV5 regulation by physiological and pathophysiological modulators

Edwin C. Fluck et al.

Summary: Transient receptor potential vanilloid 5 (TRPV5) is a calcium-selective ion channel that plays a crucial role in maintaining calcium homeostasis in the kidney. This study investigates the regulation of TRPV5 by phosphatidylinositol 4,5-bisphosphate (PI(4,5)P-2) and calmodulin (CaM), as well as the effects of parathyroid hormone (PTH), metabolic acidosis, and protein kinase A (PKA) on TRPV5 activity. The findings reveal that low pH inhibits TRPV5 by preventing PI(4,5)P-2 activation, and PKA controls TRPV5 activity by preventing CaM binding and channel inactivation.

TRPM3 in Brain (Patho)Physiology

Katharina Held et al.

Summary: Ion channels play a crucial role in regulating neural processes in the brain, and disruptions in their functions can lead to various neurological disorders. TRP ion channels are key players in the development and physiological functions of the central nervous system, influencing processes like neurite growth and synaptic plasticity. TRPM3, a non-selective cation channel, is emerging as a new player in the brain with potential roles in neuropathological processes.

PIP2 regulation of TRPC5 channel activation and desensitization

Mehek Ningoo et al.

Summary: TRPC5 ion channels, expressed in the brain and kidney, are promising therapeutic targets for diseases like focal segmental glomerular sclerosis. The role of PIP2 in controlling TRPC5 channel activity through gating rather than cell surface density, influenced by DAG and lanthanide ions, provides new opportunities for targeted therapies in chronic kidney diseases.

Dual regulation of TRPV1 channels by phosphatidylinositol via functionally distinct binding sites

Aysenur Torun Yazici et al.

Summary: Regulation of the TRPV1 channel by phosphoinositides is complex, with PtdIns potentially acting as a competitive antagonist or activator depending on the presence of PtdIns(4,5)P-2. Different binding sites for phosphoinositides may explain the varied effects observed on TRPV1 activity.

Impact of the Protonation State of Phosphatidylinositol 4,5-Bisphosphate (PIP2) on the Binding Kinetics and Thermodynamics to Transient Receptor Potential Vanilloid (TRPV5): A Milestoning Study

Arman Fathizadeh et al.

Summary: The binding of phosphatidylinositol 4,5-bisphosphate (PIP2) to the ion channel transient receptor potential vanilloid 5 (TRPV5) is critical for its function, with atomically detailed simulations and the milestoning theory used to compute the free energy profile and kinetics. The protonation state of PIP2 and the influential channel residues were identified, with the switch between protonation states modeled as a diabatic transition halfway through the reaction.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
