PPE51 mediates uptake of trehalose across the mycomembrane of Mycobacterium tuberculosis

Summary: The disaccharide trehalose is essential for the viability of Mycobacterium tuberculosis, which synthesizes trehalose de novo but can also use exogenous trehalose. The transport of trehalose and its analogues across the mycomembrane of M. tuberculosis is exclusively mediated by the PPE51 membrane protein. Mutants lacking the PPE51 gene are unable to grow on trehalose as the sole carbon source.

Summary: In acidic pH conditions, Mycobacterium tuberculosis adapts its metabolism to preferentially assimilate lipids, like oleic acid, over carbohydrates. Lack of certain enzymes necessary for lipid assimilation can be lethal to M. tuberculosis under acidic conditions, highlighting the pathogen's ability to alter its carbon diet in response to pH stress.
